Correlation Between BDNF Gene Polymorphism Andptsd Cognitive Function In Patients With Liver Cancer

Objective:Posttraumatic stress disorder(PTSD)is a psychiatric disorder that plagues trauma survivors.However,the long-term efficacy for PTSD remains controversial.Increasing evidence shows that BDNF may be involved in the occurrence of PTSD.Here we tried to demonstrate whether BDNF gene polymorphisms are correlated with neurocognitive function of PTSD in patients with hepatocellular carcinoma(HCC).Methods:This study included 102 HCC patients who complicated with PTSD,146 HCC patients and 152 healthy volunteers.Initially,we evaluated the neurocognitive function of the study subjects by mini-mental state examination.Next,We detected the polymorphisms of BDNF G11757 C and rs6265 by polymerase chain reaction-restriction fragment length polymorphism.Then,we detected serum BDNF level by means of enzyme-linked immunosorbent assay and analyzed the correlation of BDNF polymorphisms,BDNF level and HCC complicated with PTSD.Lastly,logistic regression analysis was performed.Results:The smoking history,drinking history and hepatitis B rate of HCC patients who complicated with PTSD and patients with simple liver cancer were significantly higher than those of the control group;Serum BDNF levels and MMSE scores in HCC patients who complicated with PTSD were significantly lower than those in patients with simple HCC and controls(all P<0.05).The Serum BDNF levels in liver cancer patients are positively correlated with the MMSE scores of PTSD.Haplotype GA can increase the risk of PTSD in patients with liver cancer(OR=1.521,95%CI=0.673-1.982,P=0.018),while haplotype CG can reduce the risk of PTSD in patients with liver cancer(OR=0.478,95%,CI=0.314-0.876,P=0.004).Logistic regression analysis showed that smoking history,drinking history and hepatitis B were important risk factors for HCC;low BDNF levels were an important risk factor for PTSD in HCC patients;rs6265 GG genotype was an important protective factor for PTSD in HCC patients(P< 0.05).Conclusion:Collectively,this study defines that the GG genotype of rs6265 in the BDNF gene is associated with PTSD in liver cancer and is an important protective factor for PTSD in liver cancer.Additionally these results provided a promising target for PTSD prevention in patients with HCC.
