Found in Hainan,among other places, Calophyllum inophyllum Linn [Guttifereae] of the genus Calophyllum is an evergreen tree. Widely used in folk medicine it has Anti-corrosive, convergent, anti-herpangina, analgesic,anti-inflammatory properties. C. inophyllum is well-known as a source of biologically active compounds such as Xanthonoids, Flavonoid, Coumarins, Terpenoid. Various bioactivities of antitumor-promoting, anti-cytotoxic, anti-microbial, anti- rheumatic and so on have been revealed. In 1992, the research group (U.S) reported that calanolides, isolated from C.lanigerum and C. inophyllum L.,respectively ,showed strong activity against human immunodeficiency virus(Kashman et al). Since then ,the chemical constituents of several Calophyllum species have been extensively studied. The blossoms give off a delightful, sweet fragrance.The distillation of the seeds yields a rich oil which can be used as an additive in cosmetics and other fragrant materials.A sensitive, circular, dynamic headspace sampling method was applied for collecting the volatile fragrant components of the fresh flowers of C. inophyllum L. and were identified by a combination of TCT and GC-MS. 24 components have been detected, Those components include 2-Ehtylpentane; Butane, 1,2-epoxy-3-methyl-; Butane, 1,2-epoxy-2-methyl-; Diketene; 2-Butanone, 4-hydroxy-; Methylcyclopentane; 3-Hexanol; 2-Butanol etc., which together occupied 90.28% in the total area.The concretes and essential oil from the fresh flower of C. inophyllum Linn was obtained by steam distillation ,petrol-ether and SCF. The distillations yielded 0.06%,0.33%, 0.20%, respectively. The chemical components of the oil were examined using the GC-MS technique, and the differences of the major constituents of the oil acquired by the three methods were analyzed and compared.25 compounds in the oil by steam distillation were identified. The principal constituents are Gennacrene-D (22.62%),Trans-Caryophyllene( 12.03%), β-sesquiphellandrene (19.00%). 25 compounds in the oil produced by petrol-ether were identified. Of these the principal constituents are Germacrene-D (21.24%), β-sesquiphellandrene (9.07%), Dodecatriene(5.69%). 19 compounds in the oil by SCF were identified. The principal constituents are Germacrene-D(17.70%), Trans- Caryophyllene(15.43%), kaur-16-ene (11.55%),a-Copaene( 10.62%).The essential oil from the dry seeds of C. inophyllum L. was obtained by steam distillation. The chemical components of the oil were examined by the GC-MS technique. 37compounds of the oil were characterized. 33 compounds amounting to 99.26% of the total contents were identified.The principal constituents are Dibutyl phthalate(28.80%); Palmitic acid(7.07%); Hexadecanoic acid, ethyl ester (8.05%); Oleic acid,ethyl ester(18.53%); 9,12-Octadecadienoic acid,methtyl ester(17.45%).The study on Coumarins and Xanthones derivatives from the seeds of C. inophyllum L.: The Coumarins and Xanthones derivatives were isolated based on their physical,chemical and spectroscopy properties .Several methods were used to distill and purify those ingredients, such as organic solvent distillation ,steam distillation in low pressure, chromatography , continuous crystal and so on . The IR, UV, 1H ,13C NMR, GC-MS, EIMS techniques were applied to analyze and identify the consitutents and structures of those chemical ingredients. The study has resulted in the isolation 10 chemical compounds. The structures of 5 compounds were determined by analysis ,named Calophyllolide(1), Isoino-phynone(2), Inophyllum C or Inophyllum E(4), Inocalophyllin B(5), caloxanthone E(3),and with 5 other unknown compounds. It is the first time that those 5 compounds were isolated from the seeds of C.inophyllum Linn in Hainan .
