Objective: The purpose of this study was to determine the SELDI ProteinChip technology for the automated analysis of different expression proteins between Vernal Keratoconjunctivitis and healthy subjects in tear fluid.Methods: Patients with Vernal Keratoconjunctivitis (VKC, n = 22) and healthy subjects (CTRL, n = 22) were examined. Their tear proteins were analyzed using SELDI-TOF-MS ProteinChip Arrays with NP20 ProteinChips and the map of tear protein mass spectrum was established . The data were analyzed by Ciphergen ProteinChip 3. 1. 1 software and the test of variances was done in the same M/Z protein profiles of the two groups.Result: Three different expression proteins were found between Vernal Keratoconjunctivitis and healthy subjects in tear fluid. Discriminant analysis demonstrated highly significant changes in the protein profiles in patients with VKC (P < 0.001). The molecular mass of three different expression proteins was 14950Da(P=1.94E-05),> 15147Da(P=a00013) and 3735Da(P=2.94E-05). With the protein profiles of tears in the healthy subjects, three of the peak intensity in the different expression proteins are all down obviously in the Vernal Keratoconjunctivitis.
