ObjectiveTo detect the urinary proteomic patterns of diabetic by using ProteinChip array technology , for discussing the discriminatory proteins to diagnosing early diabetic nephropathy. Methodssurface-enhancedlaser desorption / ionizationtime of flight mass spectrometry(SELDI-TOF-MS ) H4 ProteinChip was used to detect the urinary proteomic patterns of 106 patients with diabetes mellitus and 50 healthy persons. ProteinChip reader and Biomarker Wizard software were combined with a bioinformatics method to analyze protein fingerprinting. All the samples are divided into 4 groups:A group:the control group;B group: Alb (albumin) <30mg/g.cr;C group: 30300mg/g.cr .Then we obtaine some discriminatory protein peaks by group analysising . ResultsThere were over 1000 protein the protein chip obtained, about 200proteins of them were obviously different between the diabetes group and the control. Then analyzing the protein peaks by Alb partition, some discriminatory protein were found ,such as the minimus protein peaks (2197.3 m/z);the maximum protein peaks (79613.1 m/z) .The eight of them were up-regulated, and sixteen down-regulated. There are 5 small molecul protein more especial then Alb. ConclusionUsing ProteinChip technology can screen out urinary discriminatory proteins from diabetic quickly and efficiently. The urinary proteomic patterns are obviously different between early diabetic nephropathy and non nephropathy of diabetes patients, some of them come to increase with the urinary Alb increasing.
