This phenomenon urge people to find the markers related with prognosis. Our research was to detect the expression of nm23, p53, ki-67, cyclin E and their significance using immunohistochemical staining, to identify more valuable and more significant markers to direct clinical therapy and estimate prognosis.Methods:1. Construction of tissue microarray Recheck the HE tissue section of breast cancer , choose the area of more tumor cells and with no hemorrhage and necrosis, Taking out tissue cores with 2mm in diameter from donor wax lump and pushing in another wax lump to construct 1 tissue microarrays . Set a control with 13 examples to validate the effect of immunohistochemical stain. The utilizing rate of our tissue microarray is 92.3%. Choose 30 samples randomly to make full tissue sections toevaluate the reliability and feasibility of tissue microarrays.2. Immunohistochemical staining The tissue microarrays were analyzed by nm23, p53, ki-67 and cyclin E using immunohistochemical methord according to S-P protocol. Full tissue sections were stained all the same.3. Staining scoring â‘ nm23: Stains located in cytoplasm.immunohistochemical stain of tissue microarray were scored according to staining intensity; Full tissue... | | Keywords/Search Tags: | breast cancer, tissue microarray, immunohistochemistry, nm23, ki-67, p53, cyclin E | PDF Full Text Request | Related items |
