Objective: By observing the expression of pl6, cyclinDl and Ki-67 in common histological types of lung cancer, to investigate the meaning and the correlation of their expression in lung cancer.Method: Eighty samples were selected from operations during 2000-2003 in Jiangxi Provincial People's Hospital. Among them, 48 male, 32 female; Age: 39-70, average 63.4; Histological type: 37 Squamous carcinoma, 28 adenocarcinoma, 9 small-cell carcinoma, 6 large-cell carcinoma; Lymph nodal metastasis: 56 positive, 24 negative. We detected the expression of pl6, cyclinDl and Ki-67 in lung cancer, using tissue microarray technique and immunohistochemistry method. Experiment data were statistically analyzed by x~2 test, t test and linear regression.Result:The positive rate of pl6 is 58.8% in 80 cases of lung cancer, and the positive rate is 60.0% in 30 cases of biopsy withnormal or inflamematory bronchus mucosa taken during bronchofibroscopy (P>0.05).The positive rate of cyclinDl is 51.3% in 80 cases of lung cancer, but 6.7% in the 30 cases of control group (P<0.01). The proliferatin index (PI) is 62.34+21.17 in the 80 cases of lung cancer, but 8.47 ±13.64 in 30 control cases (P<0.01). The positive rate of pl6 in the 56 cases with metastasized lymph nodes is 51.8% , but 75.0% in the other 24 cases of the 80 cases of lung cancer (P<0.05). The positive rate of cyclinDl in the 56 cases with metastasized lymph nodes is 69.6% , but 8.3% in the other 24 cases of the 80 cases of lung cancer (P<0.01). PI is 73.82±14.85 in the 56 cases with metastasized lymph nodes, and 41.12±12.76 in the other 24 cases (P<0.01). Among the 47 cases of lung cancer with positive pl6, cyclinDl is expressed only in 9 (19.1%), but it is expressed in 32 cases among the 33 cases with negative pi6 (96.9%). There is significant difference between them (P<0.01). The PI in 33 cases of lung cancer with negative P16 is 74.75±15.31, and the PI is 52.18±12.47 in those with positive P16 and it is 40.62±8.81 in 12 cases with intensively expressed pl6. There is significantdifference when compare the negative ones with the positive and the negative ones with strong positive (P<0.01). The PI in 39 cases of lung cancer with negative cyclinDl is 43.26±17.34, and the PI is 73.35±13.67 in those with positive cyclinDl and it is 79.87±11.33 in 13 cases with intensively expressed cyclinDl. There is significant difference when compare the negative ones with the positive and the negative ones with the intensively positiveConclusion: The intense expression of cyclinDl is associated with the occurrence of lung cancer and metastasis of lymph nodes. But pi6 has no relation with the occurrencee of lung cancer and the histological type of lung cancer. The low expression of pl6 is closely related with lymph metastasis of lung cancer. The expression of Ki-67 is associated with the occurrence and metastasis of lung cancer, but has no relation with the histological type of lung cancer. The expression of pl6 negatively correlated with that of Ki-67, but the expression of cyclinDl has obvious positive relation with that of Ki-67.
