Objective : To explore the relationship between the expression of nm23,p53 protein and the oncogenesis development and metastasis of laryngeal cancer.Meanwhile we want to provide some objective basis for relationship among tumor suppressor gene and mechanism of laryngeal cancer and gene therapy.Method: Expressional level of n m23,p53 protein was detected in 30 human laryngeal cancer samples and 5 normal tissues adjacent to cancer respectively ,and their correlation with lymph node metastasis and the degree of cell differentiation was studied by immunohistochemistry(IHC) methods on tissue microarray . The stastitical evaluation was performed with SPSS software for windows.Results: The positive expression for nm23 in cancer samples,in human tissues adjacent to cancer,in primary cancer with lymph node metastasis and of recurrence cancer was 46.67%(14/30), 100%(5/5)), 23.07(3/13),18.18(2/11) respectively . The positive expression for p53 was 56.67%(17/30), 0(0/5), 84.61%(10/13),90.91%(10/11) respectively . The results showed a significant association between P53 positive expression and cervical node metastasis.(P<0 .05).The nm23 negative expression had a significant association with cervical lymph node metastasis( P<0 .05).The expression level of nm23 and p53 were closely related to the degree of cell differentiation(P<0 .05) and cancer recurrence (P<0 .05).In addition, the expression level of nm23 and p53 gene was related to the clinical period of laryngeal cancer(P<0 .05).But nm23 and p53 gene was not related to the clinical type of laryngeal cancer ( P>0 .05).Conclusions: These results suggest that nm23 gene and p53 gene are closely related to the clinical period of laryngeal cancer, the degree of cell differentiation ,lymph node metastasis and recurrence of laryngeal cancer .The lower expression level of nm23 and the higher expression of p53 predicts the metastasis and recurrence of laryngeal cancer,and the two genes may be an index in monitoring the status and evaluating the prognosis of the lung cancer.
