The sufferer is in high risk of suicide crowd,from the period of taken bad to recover,always suffer the dangerous of suicide. The incidence and sicken rate of MD are very high as the evolution of MD, it's still enhance the rate of other disease, make the sufferer's existence rate and quality of life consumedly descend, and created variety negative influence to society and family .The research of MD has became a very important topic that having been caused science foundation of the spirit nerve and clinical workers'biggest concern, currently, some research relevant MD have made substantial progress.The science of traditional Chinese medical(TCM) have made more and more exertive in the MD medical treatment, the treatment of MD should follow the dialectics of dumps as a cure method, consider the mechanism of the disease is uncomfortable in feeling ,cause block in the function of liver, and gradually with the result that the other five internal organs in a relation of disorder ,and Jian Pi Shu Gan was exactly a efficiency therapy in MD treating principle. Through the research to the theories and clinical result of TCM and modern medical, developed curative effect, lower in side-effect Chinese traditional medicine to anti-MD, that has expand the science of TCM culture, enhance TCM of clinical application, and push TCM modernization, have graveness of theories and experiment meaning .Professor Li Youtian in the traditional medical department of the first hospital of Jin Lin university developed Chinese herbal medicine named Shugan Jieyuyin Self-made (SJSM) that mainly used for treating MD has been approval the product inside the hospital. It may relieve the liver pain, safeguard stomach against damages and soothe the nerve and so on. The clinical curative effect has got first step confirmation, but the influence of this prescription on blood serum LPO and experiment research haven't appeared in report, so through this experiment, the research of SJSM in treating depression model mice on blood serum LPO, then provide a pharmacology foundation for clinical treatment in MD.Aim: Adopt isolated feed and CUMS that is currently extensive application by domestic and international; imitate to result in depression model mice. Measure the mice weight and psychosis changes by week, measured the consumption of 1% sucrose solution,using Open-field experiment combine slightly changes to measurement the mice behavior to get the account, and make use of reagent boxes with the method of showing a color to measure the activity of serum SOD and serum NO,MDA levels in depression model mice. Observation on the level of blood serum LPO of depression model mice, and the influence of SJSM to LPO of depression model mice, then study the main mechanism of function for the development of SJSM and provide a credibility basis for clinical usage.Material and method:chose 48 healthy wistar mice, all male, divided into a normal group, model group, medicine group including big,medium and small, Bai Youjie group at random, all fed isolated. In addition to normal group, everyday give rest group big mice one kind of stimulation include: fasting for 24hours, force swimming, electric shock foot, hang tail, forbid water, inclined cage for 24hours and day and night reverse for 24hours, 7days is a circulation, total 56 days. At the 28th day, beginning to medicine the mice through the method of infuse stomach, 2ml everyday, totally 28 days, dealt normal group and model group with equal physiology brine, dealt medicine big,medium and small group with the medicine density of 16 g/ml,8 g/ml and 4 g/ml, dealt Bai Youjie group with 0.18 g/ml Bai Youjie. Measured the mice weight and psychosis changes by week, measured the consumption of 1% sucrose solution every two weeks,and measured the mice behavior and got the account at the 28th and the56th day. At the 56th day of experiment, used aether to make the mice in narcosis, then through the method of extirpate eyeball to got blood 3-5ml, used centrifugal machine to obtained blood serum. Make use of reagent boxes with the method of showing a color measured the activity of serum SOD and serum NO,MDA levels in depression model mice. Observed on the level of blood serum LPO of depression model mice, and the influence of SJSM to LPO of depression model mice.Result: compare with the normal group, every groups of mice been stimulated, the increment of weight number decrease obviously, the mental state and behavior all have changed, with the performance of fear and sense of incapability, lower reflection toward to outside stimulate. At the 28th day, by the method of Open-field, every groups of mice been stimulated have score little than normal group(P<0.01),the weight number is less than normal group(P<0.01),and the consumption of 1% sucrose solution is less than normal group(P<0.01)too;by using three dosage of SJSM and Bai Youjie infusing stomach treatment for 28 days, the medicine big and medium groups'weight number is more than normal group(P<0.01) or(P<0.05), they have score more than the model group(P<0.01) or(P<0.05),and the consumption of 1% sucrose solution is more than model group(P<0.01);After treated, by using of reagent boxes with the method of showing a color, compare with the model group, the activity of serum SOD raised obviously, and the serum NO,MDA levels declined obviously in every treatment group(P<0.01 or P<0.05),and the activity of serum SOD and serum NO,MDA levels were equal to the normal group, it showed that the effect of Chinese medicine was superior to Western medicine in some group.Conclusion:1,CUMS can make the big mice in depress appearance, and copy out depression model mice successfully.2,SJSM can resume the increment of weight number, increase the degree of big mice behavior, increase the consumption of 1% sucrose solution and have the effect on improve the state of depression mice.3,The activity of serum SOD raise obviously and the serum NO,MDA levels decline obviously in the depression model mice, maybe it is concerned with the occurrence of MD, and it support hypothesis theory that the oxygen free radicals has transform in MD sufferer. 4,SJSM can rise the activity of serum SOD and reduce the serum NO,MDA levels in the depression model mice.5,The result of the experiment clue to the changes in LPO was possibility related to the mechanism of MD, and clue to the mechanism of SJSM anti-MD from serum LPO side, provide a pharmacology foundation of SJSM for clinical usage.
