Objective:To investigate the expression of HIF--1αand VEGF in cervical tissue and their correlation with the Surgical and Pathology Assessment.pathological grade and lymph node metastasis of endometrial adenocarcinoma of uterus. To estimate the state and prognosis of the patient and to find good molucular markers and a new way to therapy the carcinoma.Methods:1.The expression of HIF--1αand VEGF in 52 cases of endometrial adenocarcinoma of uterus and 20 cases of normal endometrial tissue were assessed by immunohistochemical staining2. 30 cases of fresh endometrial adenocarcinoma of uterus and 12 cases of normal endometrial tissue were determined by westen-blot method.3. Using SPSS 15.0 software to assay the data.Rusults:1. Immunohistoc hemical staining : (1)HIF-1αwas staining in the neclear or plasma , HIF-1αwas expressed in normal endometrium during menstrual cycle (including proliferative and secretory phase ). HIF-1αexpression in proliferative phase(positive staining rate 30%) were higher than in secretory phase ,but lower than in endometrial adenocarcinoma of uterus(positive staining rate 65.38%) .( Z=-5.162, P < 0.01). The expression of HIF-1αelevitated as the pathologic grade decreased (Z=-2.769, P<0.01) and the surgical and pathology assessment increased ( X2=15.450, P<0.01). the expressing of HIF-1αhas no correlation with lymph node metastasis (Z=-1.77,P>0.05 ).(2) VEGF was staining only in the plasma ,VEGF was expressed both in endometrial adenocarcinoma of uterus(63.46%) and nomal endometrial tissue(40%),.VEGF level inendometrial adenocarcinoma of uterus were much higher than those in nomal endometrial tissue, (Z=-5.126, P<0.01) The expression of VEGF elevitated as the pathologic grade decreased (Z=-3.528 ,P<0.01) and the surgical and pathology assessment increased (X2=13.318,P<0.01). the expressing of VEGF has no correlation with lymph node metastasis (Z=-0.515,P>0.05 ) (3)VEGF has positive correlation with HIF-1α2.Western-blot: the protein band of HIF-1αand VEGF is at 120KD and 35KD,using UVP system to take picture and note the OD. The OD of HIF-1αis (3.84±1.33)times while VEGF is (4.91±1.26) times in endometrial adenocarcinoma as that in normal proliferative endometrial tissue.Conclusions:1.HIF-1αwas staining in the neclear or plasma , HIF-1αwere both expressed in normal endometrium during menstrual cycle (including proliferative and secretory phase ) and in endometrial adenocarcinoma of uterus. VEGF was staining only in the plasma ,VEGF was expressed both in endometrial adenocarcinoma of uterus and nomal endometrial tissue. It is determined that HIF-1αand VEGF expression in proliferative phase were higher than in secretory phase ,but lower than in endometrial adenocarcinoma of uterus .2. The expression of HIF-1αand VEGF in endometrial adenocarcinoma of uterus elevitated as the surgical and pathology assessment increased , the pathologic grade decreased . HIF-1αand VEGF are good molucular markers to endometrial adenocarcinoma,which can be used to assist diagnosis and predict prognosis.3. The expression of HIF-1αand VEGF have no correlation with lymph node metastasis ,which can't be used to judge lymph node metastasis in endometrial adenocarcinoma.4. VEGF has positive correlation with HIF-1α. HIF-1αmay be one of the important regulator of VEGF.5.The expression of HIF-1αand VEGF can be used to accurately assess the prognosis in endometrial adenocarcinoma of uterus, further understanding of HIF-1αmay provide new therapy regimens for endometrial adenocarcinoma of uterus.
