Objective To investigate the expression of HIF-1αand VEGF in ovarian tissue and their correlation with the clinicalstage and pathological grade of ovarian cancer. To estimate the state and prognosis of the patient and to find good molecular markers and a new way to therapy the cancer.Methods Chosing 58 cases of ovarian cancer tissue and 23 cases of borderline tumor tissue as test group. Distinguishing by clinical stage ( FIGO standard ) : stsgeⅠ8 cases; stageⅡ13 cases; stageⅢ29 cases and stageⅣ8 cases. Distinguishing by tissue grade (WHO standard) : well to moderately differentiated 9 cases ; poorly differentiated 49 case ovarian tissue as control group. The expression of HIF-1αand VEGF were assessed by immunohistoc hemical staining and western-blot method in ovarian cancer tissue , borderline tumor tissue and nomal ovarian tissue. Using SPSS 12.0 software to assay the data.Results Immunohistochemical staining : HIF-1αwas staining in the nuclear or plasma , HIF-1αpositive staining rate is 86.21% in ovarian cancer , 47.83% in borderline tumor tissue and 13.16% in normal ovarian tissue. The difference of the three groups was statistically significance . The expression of HIF-1αelevitated as the clinical stage increased(P<0.05) and the pathologic grade decreased(P<0.05). VEGF was staining only in the plasma . VEGF was expressed in nomal ovarian tissue , borderline tumor tissue and ovarian cancer , the rate of positive expression was 57.89%,82.61%,98.28% respectively. Deferrence was seen among three groups (P<0.05). VEGF level in ovarian cancer was much higer than those in nomal ovarian tissue, The expression of HIF-1αelevitated as the clinical stage increased(P<0.05)and the pathologic grade decreased(P<0.05). VEGF has positive correlation with HIF-1α. Western-blot: the protein band of HIF-1αand VEGF is at 135KD and 23KD, using UVP system to take picture and note the OD. The OD of HIF-1αis (5.1±1.62)times while VEGF is (4.2±1.62) times in ovarian cancer as that in normal ovarian tissue.Conclusions1.HIF-1αwas staining in the neclear or plasma , HIF-1αpositive staining rate is 86.21% in ovarian cancer , VEGF was staining only in the plasma , VEGF was expressed in nomal ovarian tissue , borderline tumor tissue and ovarian cancer , positive staining rate is 98.28% in ovarian cancer.2.The expression of HIF-1αand VEGF in ovarian cancer is much higher than nomal ovarian tissue.3.The expression of HIF-1αand VEGF elevitated as the clinical stage increased and the pathologic grade decreased.4.VEGF has positive correlation with HIF-1α.5.The expression of HIF-1αand VEGF can be used to accurately assess the prognosis in ovarian cancer, further understanding of HIF-1αand VEGF may provide new therapy regimens for ovarian cancer and pathogenesis...
