Objective To investigate the expression of HIF-1αand VEGF in cervical tissue and their correlation with the clinical stage,pathological grade, invasion deepth and lymph node metastasis of cervical squamous cell carcinoma. To study the correlation between HIF-1αand VEGF .To estimate the state and prognosis of the patient , to study the mechanism of cervical squamous cell carcinoma and to find a new way to therapy the carcinoma. Methods 1. The expression of HIF-1αand VEGF in 62 cases of cervical squamous cell carcinoma and 11 cases of nomal cervical tissue. were assessed by immunohistoc hemical staining . 2. 30 cases of fresh cervical squamous cell carcinoma and 11cases of nomal cervical tissue were determined by western-blot method. 3. Using SPSS 10.0 software to assay the data. Rusults 1. Immunohistoc hemical staining : (1)HIF-1αwas staining in the neclear or plasma , HIF-1αwas only expressed in cervical carcinoma, while there is no expressing of HIF-1αin normal tissue or carcinoma in situ. HIF-1αlevel in cervical carcinoma were much higer than those in nomal cervical tissue, the expression of HIF-1αelevitated as the clinical stage increased ( X~2=32.082,P<0.01 ) and the pathologic grade decreased ( Z=-3.855 ,P<0.01 ). Stat. deferrence was seen among carcinoma in situ , early invasive stage and lately invasive stage ( X~2=22.881,P<0.01) .the expressing of HIF-1αhas no correlation with lymph node metastasis ( Z=-1.175 ,P>0.05 ) .(2) VEGF was staining only in the plasma ,VEGF was expressed both in cervical squamous cell carcinoma and nomal cervical tissue.VEGF level in cervical carcinoma were much higer than those in nomal cervical tissue, the expression of HIF-1αelevitated as the clinical stage increased ( X2=19.660,P<0.01 ) and the pathologic grade decreased ( Z=-4.245 ,P<0.01 ). Stat. deferrence was seen among carcinoma in situ , early invasive stage and lately invasive stage ( X~2=15.988,P<0.01). The expressing of HIF-1αhas no correlation with lymph node metastasis ( Z=-1.175 ,P>0.05 ) .(3)VEGF has positive correlation with HIF-1α. 2. Western-blot: the protein band of HIF-1αand VEGF is at 120KD and 23KD,using UVP system to take picture and note the OD. The OD of HIF-1αis (5.1±1.62)times while VEGF is (4.2±1.62) times in early invasive stage as that in normal tissue. Conclusions 1. HIF-1αwas stained in the neclear or plasma , HIF-1αwas only expressed in cervical carcinoma, while there is no expressing of HIF-1αin normal tissue or carcinoma in situ. VEGF was stained only in the plasma ,VEGF was expressed both in cervical squamous cell carcinoma and nomal cervical tissue.It's determined by immunohistoc hemical staining and western-blot that the expression of HIF-1αand VEGF in cervical squamous cell carcinoma was much higher than that in nomal cervical tissue. 2. The expression of HIF-1αand VEGF in cervical squamous cell carcinoma elevitated as the clinical stage increased, the pathologic grade decreased and invasive depth added. HIF-1 αand VEGF are goodmolucular markers to cervical squamous cell carcinoma,which can be used to assist diagnosis and predict prognosis. 3. The expression of HIF-1αand VEGF have no correlation with lymph node metastasis,which can't be used to judge lymph node metastasis in cervical squamous cell carcinoma. 4. VEGF has positive correlation with HIF-1α. HIF-1αmay be one of the important regulator of VEGF. 5. The expression of HIF-1αand VEGF can be used to accurately assess the prognosis in cervical squamous cell carcinoma, further understanding of HIF-1αmay provide new therapy regimens fo cervical cancer.
