Objective To study the expression and correlation of COX-2 and MMP-2 in rectal cancer.Methods We have collected 68 cases of rectal cancer from Oct,2002 to Oct,2005 which have been proved in pathology. Among them male 38, female 30, mean age 57.3,and 27 cases were well-differentiated,30 were moderately differentiated, 11 were poorly differentiated. With lymphatic metastasis were 40 cases and 28 cases without lymphatic metastasis. 15 cases had reccurence after operation, and 53 cases without recurrence.29 cases belonged to Dukes A and B stage ,and 19 belonged to Dukes C and D stage . 20 normal large intestine mucous membrane were taken as the contrast group.Immunohistochemistry staining(Streptavidin-Peroxidase /S-P method) was performed to detect the expression of COX-2 protein and MMP-2 protein. The criteria of positive expression was that brown granule was found in cytoplasm.Cytes of COX-2 or MMP-2 positive expression in ten high power field were counted randomly in each case. (1) Positive(+): positive cytes account 10%~30%;(2) positive (++): positive cytes account 30~50%,(3) positive (+++): positive cytes account >50%; negative (-):positive cytes account <10%. The data were analyzed by software SPSS 10.0. The difference of COX-2 and MMP-2 expression was analyzed with Chi-square Test and exact probability method. The difference of COX-2 and MMP-2 expression among different pathological classifications of rectal cancer was analyzed in the same way.
