| Objective:The expression of IER3 and circadian protein in rectal cancer was detected.The relationship between the expression of IER3 and clinicopathological parameters and prognosis was analyzed statistically.Objective to investigate the clinical significance and value of IER3 in rectal cancer.Methods:(1)Using immunohistochemistry,The expression of IER3 in 42 rectal cancer and adjacent tissues was detected;(2)the mRNA of IER3 in 42 cases of rectal cancer and normal tissues was detected by quantitative real-time PCR(qRT-PCR)The expression level of the tumor cells was detected and classified in 42 patients with rectal cancer during perioperative period,and a new type of multiple mRNA in situ analysis(MRIA)was used,method was used to detect the mRNA expression of IER3 in the enriched CTC cells;(4)the relationship between the expression of IER3 gene and the clinicopathological characteristics of colorectal cancer and peripheral blood circulation tumor cells was analyzed statistically.Kaplan Meier method was used to analyze the relationship between the expression of IER3 gene and prognosis of rectal cancer patients.The difference between log rank groups was compared.Results:(1)The expression of IER3 in rectal cancer tissues was higher than that in adjacent normal tissues(92.9% VS 14.3%,X2=49.012,P<0.001).(2)qRT-PCR results showed that the expression of IER3 mRNA in rectal cancer tissues was significantly higher than that in corresponding adjacent normal tissues(t = 2.400,P < 0.05).(3)There was no significant difference in IER3 expression between the two groups(P > 0.05).(4)The difference of IER3 mRNA expression was positively correlated with tumor size(P < 0.05);There was no significant difference between the two groups(P > 0.05).(5)There was a correlation between mixed CTCs and Ki67 positive rate before operation(P < 0.05);It was correlated with preoperative CEA level(P < 0.05).(6)Postoperative epithelial CTCs were correlated with gender,depth of invasion,TNM stage and vascular invasion(P < 0.05).(7)The expression of IER3 mRNA in preoperative mixed CTCs was correlated with preoperative CEA level(P <0.05).(8)The expression of IER3 protein was correlated with preoperative mixed CTCs and preoperative E-type CTCs(P < 0.05).(9)Survival analysis showed that there was no correlation between IER3 protein expression,IER3 mRNA expression,gender,age,tumor size,depth of invasion,lymph node metastasis,TNM stage,vascular invasion,neural invasion and prognosis of rectal cancer patients(P > 0.05).Conclusion:(1)The expression of IER3 mRNA in rectal cancer was significantly higher than that in normal tissues.(2)Preoperative mixed CTCs were correlated with Ki67 positive rate and preoperative CEA level;postoperative epithelial CTCs were correlated with gender,depth of invasion,TNM stage and vascular invasion;The expression of IER3 mRNA in preoperative mixed CTCs was correlated with preoperative CEA level.It is helpful to evaluate the invasion ability of patients.(3)The expression of IER3 protein was correlated with preoperative mixed type CTCs and preoperative E-type CTCs.IER3 was related to the occurrence and development of rectal cancer.IER3 is expected to be a biomarker of rectal cancer. |
