Osteoporosis is a kind of systemic skeletal disease characterized by low bone mass and micro architectural deterioration of bone tissues, with a consequent increase in the bone fragility and the susceptibility to the fracture. The elderly and postmenopausal women were susceptible to suffer from osteoporosis that may lead to the compressive fractures of lumbar vertebrae and the femoral neck fractures, which affect the existent qualities of the patients to a great extent. The studies of Osteoporosis are very numerous and popular, which they make some progress rapidly and deeply. Some prior investigations indicated that lack of estrogenic hormone and calcium intake less have already become the principal reason to lead to bone mass lose and osteoporosis occur. However, with the development of cryobiology and molecular biology increasingly and deeply in recent ten years, the effect and function of cytokine have been pain more and more attention, especially TNF-aandTGF-β1,which play very important role in osteoporosis. JIANGGUIIHAO was mainly basis on JIANGGULING and combined with several years clinical experiences by tutor. According to former observations and correlated empirical study, it has been gotten the positive and affirmative curative effect by using JIANGGUIIHAO. This experiment from point of immunohistochemical view aims at to study the mechanism of the treatment with JIANGUHAO on osteoporosis.To investigate the mechanism of the treatment with JIANGGUIIHAO on postmenopausal osteoporosis in vivo, adult ovariectomied rats were used in this study as the model of postmenopausal osteoporosis. The numbers of osteoblast in thebone tissues were analyzed using HE staining, and tumor necrosis factor-oc and transforminggrowthfactor-pi expressions in bone tissues of ovariectomy(OVX),ovariectomy with diethylstilbestrol, ovariectomy with JIANGUIIHAO rats were analyzed at molecular level by means from immunohistochemical observation.The results indicated that the numbers of osteoblasts of OVX-JIANGUIIHAO were more than those of OVX (p<0.01). And thetissuesshowedthatTNF-aandTGF-Piweremainlyexpressedinosteo blasts.Theresultsofimmunohistochemicaltissueshowed that the expressions of TNF-a were lower than those of VOX in the bone tissues (p<0.01), and expressions of TNF-a were higher than those of VOX in the bone tissues (p<0.05). These datum indicated JIANGULINGplayedanimportantroleintreatmentofosteoporosisbycal led"estrogen-like"effection.ItrestrictedtheTNF-acompositionandext ernalization in order to degrade its activity, at the same time, removed its depressant to the expression of TGF-Pp The result indicated that it made the activity of bone resumption on orthoclase to weaken and increased the bone formation of orthoclastic. It changed from the negative state of equilibrium of bone metabolism to positive state.
