The Experimental Study On Effects Of Anti-gout Of DGNT Pill

Objective: To study the influence of the DGNT pill on anti-inflammatory and analgesia diuresis and hyperuricemia model.Method: Establishing models of hyperuricemia by inject 300mg/kg Oteracil potassium ( OXO ) through abdomen to simulate the uric acid increase in little mice and by oral administrating 200mg/kg Adenine and 250mg/kg Ethambutol to cause mice hyperuricemia model, in which uric acid in blood serum can be observed. Acute urarthritis model can be caused by MSU in rabbit, and the influence of drug on IL-lβ, IL-8, TNF-α in joint fluid and 6-K-PGF_1α in arthral tissue was observed. At the same time, model of acute rat paw edema can be made by MSU to explore influence of anti-inflammatory. In addition the analgesic effect was observe by heat-plate and acetic acid distortion. And at the same time urine volume of mice can be observed.Results: DGNT pill have significant difference in decreasing uric acid, urea nitrogen, inosine, xanthine oxidase in serum. DGNT pill have significant difference in decreasing IL-1β, IL-8, TNF-α in joint fluid and 6-K-PGF_1α in arthral tissue. Meanwhile, pathological change of nephridial tissue be ameliorated after administration. DGNT pill can obviously ease foot swell,reduce counts of distortion and at the same time have some influence in latency period of threshold of feeling, compared with the blank model group, urine volume of mice added in group of DGNT pill significantly, but pH value has no clear change.Conclusion: the results demonstrate that the DGNT pill plays an important role in decreasing uric acid in serum and controlling the spasm of urarthritis and defending the outbreak of gouty kidney. And at the same time, the DGNT pill be well in anti-inflammatory, analgesic and diuresis.
