Experimental Study Of Angiogenesis Effects In Ischemic Myocardium Of Infarcted Rats And Its Molecular Mechanisms Induced By Yiqihuoxue Prescription

Objectives. It was studied whether Yiqihuoxue prescription have the effects of Therapeutic Angiogenesis in myocardium of infarcted rats. And we have observed the pathological and morphologic changes and the expression of CD34, VEGF-B, bFGF, PDGF-B and IGF-1 at protein level in myocardium of infarcted rats to study myocardial angiogenesis and its molecular mechanisms.Methods. The root of left coronary artery was ligated to establish heart failure after myocardial infarction in wistar rats, and then it were divided into 5 groups at random and treatmed 6 weeks with different therapy. There are the large and small doses of Yiqihuoxue Prescription group, Shexiang Baoxin Pills group, the model group, the sham operation group. Six weeks later, we observed their general status and LVMI. Then we studied the expression of CD34, VEGF-B, bFGF, PDGF-B and IGF-1 at protein level in immune histochemical method, and measured Microvascular vascular count (MVC), Microvascular vascular density (MVD) in the edge of infarct section to determine if Yiqihuoxue prescription could induce myocardial angiogenesis and evaluate its molecular mechanisms.Results. (1) Comparing of general status of rats. The mortality rate in the large dose group is more remarkably decreased than in the model group (P<0.05) . The other medicine groups had a trend in decreasing, but it is not significant than the model group (P>0.05) . Heart rate of Yiqihuoxue prescription group was reduced clearly after treated 6 weeks. And it is significant to compared with the model group (P<0.05) . (2) Comparing of LVMI. LVMI of Yiqihuoxue prescription group decrease evidently after treated 6 weeks.
