| Objective:This research adopts the ligation rat heart left anterior descending coronary artery method of copying animal model of heart failure after acute myocardial infarction(AMI),through the observation on the monkshood red halloysite pill party heart failure after acute myocardial infarction rats serum inflammatory factor and LC3 Ⅰ / Ⅱ,Beclin1,nf-kappa B signaling pathway to explore the influence of such indicators system of the degree of myocardial injury in rats with acute heart failure after myocardial infarction to improve and inhibition of ventricular remodeling,which will provide a scientific basis for clinical treatment of heart failure after acute myocardial infarction.Methods: 60 SPF male SD rats were randomly divided into normal group,model group,Wutou Chishhizhi pill low-medium-high dose group,and Qiliqiangxin capsule group,with 10 rats in each group.Except for the normal group,the model of heart failure after acute myocardial infarction was prepared by ligation of the left anterior descending coronary artery in other groups,and the elevation of the arched ST-segment was the sign of successful modeling.After 14 days of treatment,the pathomorphological changes of myocardial tissue were observed by hematoxylin-eosin(HE)staining.Masson staining was used to observe myocardial fibrosis in rats.ELISA kits in the rat serum interleukin 6(IL-6),1 beta interleukin(IL-1 beta),tumor necrosis factor alpha(TNF alpha),c-reactive protein(CRP),brain natriuretic peptide(BNP)and angiotensin Ⅱ(Ang-Ⅱ)content;The expressions of gap junction 43(CX43),matrix metalloproteinase 9(MMP9)and galactinin 3(Gal-3)in rat cardiac tissue were detected by immunohistochemistry(IHC).Western blot method to detect the rat myocardial CX43,MMP9,Gal-3 protein expression,as well as LC3 Ⅰ / Ⅱ,Beclin1,nf-kappa B protein expression levels.Results:1.Effect of Wutou chishizhi pills on pathological morphology of myocardial tissue in rats with heart failure after acute myocardial infarctionHematoxylin-Eosin Staining showed that the myocardial fibers in the normal group were neatly arranged and the cells without distortion were uniformly stained.In the model group,muscle fibers were twisted and broken,uneven cell staining,interstitial edema and inflammatory cell infiltration were observed.The arrangement of myocardial fibers tended to be orderly,interstitial edema was reduced,and inflammatory cell infiltration was reduced in the low,medium and high dose groups of Wutou chishizhi pills and Qiliqiangxin capsules.2.Effect of Wutou chishizhi pills on myocardial fibrosis in rats with heart failure after acute myocardial infarctionMasson staining showed that cardiomyocytes in normal group showed uniform red color without blue collagen fiber accumulation.In model group,a large number of blue collagen fibers were accumulated,and the arrangement of myocardial fibers was loose and disordered.The accumulation of collagen fibers in the low,middle and high dose groups of Wutou Chishhizhi pills and Qiliqiangxin capsules was improved to different degrees,and the improvement was the most obvious in the middle and high dose groups.The myocardial fibers were arranged neatly without distortion and fracture.3.Wutou chishizhi pills on heart failure after acute myocardial infarction(AMI)rats serum IL-6,IL-1β,TNF-α and CRP,the BNP,Ang-Ⅱ effectCompared with normal group,the contents of IL-1β,IL-6,TNF-α,CRP and BNP in model group were increased(P < 0.05);Compared with model group,Wutou chishizhi pills for low and middle dose group and stilbene Li strong heart capsule group of beta,IL-6,IL-1 TNF alpha,CRP,BNP,Ang-Ⅱ were reduced(P < 0.05).4.Effects of Wutou chishizhi pills on the protein expression levels of CX43,MMP9,Gal-3 and P50 in myocardial tissue of rats with heart failure after acute myocardial infarctionImmunofluorescence staining showed that compared with the normal group,the expression of CX43 protein decreased and the expression of Gal-3 and MMP9 protein increased in the myocardial tissue of the model group.Compared with the model group,the expression of CX43 protein in Wutou Chishhizhi pill low,middle and high dose groups and Qiliqiangxin capsule groups increased in different degrees,while the expression of Gal-3 and MMP9 protein in high dose group and Qiliqiangxin capsule group decreased in different degrees,while the expression of MMP9 protein in high dose group decreased significantly.5.Wutou Chishhizhi pill on heart failure after acute myocardial infarction(ami)rats LC3 Ⅰ / Ⅱ,Beclin1,protein involved in the nf-kappa B signaling pathwayWestern blot analysis showed that compared with normal group,the expression of CX43 protein was decreased and the expression of MMP9 protein was increased in model group.Compared with the model group,the expression level of CX43 was significantly increased and the expression level of MMP9 was decreased in the high-dose group.Compared with the normal group,the expression levels of LC3 and Beclin1 protein in model group were decreased,and the expression levels of p65 and IκB were decreased.Compared with model group,the expression levels of LC3 and Beclin1 in Wutou Chishhizhi pill high-dose group and Qiliqiangxin capsule group were increased,and the expression levels of p65 and IκB were increased.Conclusion: Wutou Chishhizhi pill can improve the degree of myocardial fibrosis after acute myocardial infarction(heart failure rats and delay the occurrence of ventricular remodeling and its mechanism may be Wutou Chishhizhi pill party through the lower levels of serum inflammatory factors to improve the inflammatory injury,increase the CX43 protein content in myocardial tissue,activate autophagy and the nf-kappa B signaling pathway to reduce Gal-3,MMP9 protein content in myocardial tissue and protect myocardial cells. |
