Effect Of Yiqihuoxue Prescription On Expressions Of MMP-2 And It's Influence Of Factors In Myocardial Extracellular Matrix In Rats After Myocardial Infarction

Object. In the experimental investigationIt,we have observed the pathological and morphologic changes and the expression of matrix metallopro- teinasse-2 (MMP-2),it's regulation-Tissue inhibitor of metalloproteinases (TIMP-2) and it's influence of factors ( TGF-β1,TNF-α,IL-1β) in myocardium of infarcted rats to study the effects of Left Ventricular Remodeling and its molecular mechanisms.Method. The root of left coronary artery was ligated to establish models of left ventricular remodeling after myocardial infarction in male wistar rats, and then it were divided into 5 groups at random and treatmed 6 weeks with different therapy. There are the large and small doses of Yiqihuoxue Prescription group, the control group, the model group, the sham operation group. Six weeks later,we observed their general status and LVMI. Then we studied the expression of MMP-2,TIMP-2 and TGF-β1 at protein level in immune histochemical method, and the level of TNF-αand IL-1βin the serum in radioimmunoassay assay method.Results. (1) Comparing of general status of rats. Heart rate of Yiqihuoxue prescrip- tion group was reduced clearly after treated 6 weeks. And it is significant to compared with the model group ( P<0.05 ); (2) Comparing of LVMI. LVMI of Yiqihuoxue prescription group decrease evidently after treated 6 weeks. And it is significant compared with the model group( P<0.05 ). (3) Comparing of MMP-2. Yiqihuoxue prescription group could reduce the expressions of MMP-2, it is significant compared with the model group ( P<0.05 ),and the large group is no meaning compared with the control one( P>0.05 ), there is significant meaning between the large dose and smoll dose group( P<0.05 ) . (4) Comparing with TIMP-2. Yiqihuoxue prescription group could increase the expressions of MMP-2, it is significant compared with the model group( P<0.05 ),and the large group is no meaning compared with the control one( P>0.05 ), there is significant meaning between the large and smoll group(P < 0.05) . (5) Comparing with TGF-β1. Yiqihuoxue prescription group could increase the expressions of TGF-β1, it is significant compared with the model group( P<0.05 ),and the large group is no meaning compared with the control one( P>0.05 ), there is significant meaning between the large and smoll group( P<0.05 ) . (6) Comparing with IL-1β,TNF-α. Yiqihuoxue prescription group could reduce the expressions of MMP-2, it is significant compared with the model group( P<0.05 ),and the large group is no meaning compared with the control one( P>0.05 ), there is significant meaning between the large and smoll group( P<0.05 ) .Conclusions. Yiqihuoxue Prescription, The large dose group, can reverse Left Ventricular remodeling after myocardial infarction. It was proved that the mechanisms of preventing Left Ventricular Remodeling of infarcted rats is related to reducing MMP-2 and increase TIMP-2; the mechanisms of reducing MMP-2 and increase TIMP-2 is related to reducing IL-1β,TNF-αand increase TGF-β1.
