Objective:Observing the change of wheal and flare inhibited by histamine-skin experiment through take soup of Xiaoqinglong decoction and loratadine, To compare antihistamince funtion of Chinese herb medicine and Modern medicine , and set up simple and effective drug effect dynamics parameter based on this , lay the foundation for the research for the future. Method:A total of 52 volunteers from the outpatients department were randomly divided into three groups : loratadine group, placebo group and Xiaoqinglong decoction group. Histamine-skin experiment(histamine concentrations 20.1, 7. 3, 2. 7andl. 0 mraol/L) were performed for each one before dosing and 2, 5 and 24 hours after dosing at forearm. The change of wheal and flare areas was measured after 15 minutes, draw the area under dose -response curve (AUDRC) and calculated them. ( 2) Count areas and carry on multifactor plural analysis of variance to all groups of data with SPSS10.0, according to the result, compare antihistamine function of Chinese herb medicine and Modern medicine, and set up the dynamics model of drug effect. Result:1.The wheal and flare area compared with itself after dosing and before dosing : There was no significant statistical difference between Xiaoqinglong decoction and the loratadine group(P>0. 05 );2. The wheal and flare area compared each among three before dosing and 2^ 5 and 24 hours after dosingNo significant statistical difference among Xiaoqinglong decoction, the loratadine group and plaebo group before dosing (P> 0.05 ), no difference significance between Xiaoqinglong decoction group and placebo group at 2, 5, and 24 hours after dosing (P> 0. 05 ), but the loratadine group compared with placebo group, difference have significance ( P<0. 01) very highly. 3 AUDRCThe dose-response curves was no significant difference in Xiaoqinglong decoction group and placebo group. AUDRC have no significant difference among in Xiaoqinglong decoction group , loratadine group and placebo group beforedosing. No difference significance between Xiaoqinglong decoction group and placebo group at 2, 5, and 24 hours after dosing (P> 0. 05 ), The AUDRC of loratadine compared with with placebo shows a highly significant. ( P<0.01) Conclusion:1. According to restraint function in histamine-skin-test of the antihistamine medicine can set up the dynamics model of drug effect . AUDRC can be as an index that compare antihistamine medicine efficacy of medicine dynamics activity.2. Before dosing and after dosing , the wheal and flare area and AUDRC did not have significance change by taking Xiaoqinglong decoction, It proved that Xiaoqinglong decoction cannot inhibit the histamine directly , but might inhibit Mast cells from degranulation , reduce the secretion of the histamin .3. We infer that antihistamince funtion of Chinese medicine and Modern medicine are: the same one is the both can't inhibit histamine directly, Modern medicine effected through fighting for histamin recipient of cell membrane with histamin , but, inhibit the histamin indirectly. Traditional Chinese medicine inhibit Mast cells from degranulation, reduce the secretion of the histamine.
