| Objective To explore the intervention of Cordycepssinensis on TGF- β1 of the progressive glomerulosclerosis in rats,and discuss mechanism of action.Methods 36 healthy 50-day-old Wistar rats were equally divided into three groups at random: normal control group, progressive glomerularsclerosis group made by 5/6 nephrectomy and Cordycepssinensis -treated progressive glomerularsclerosis group after the operation of 5/6 nephrectomy for 4 weeks or 9 weeks. Water were poured down into the stomach of rats in the group of the progressive glomerularsclerosis and normal control every day.4 weeks later six rats in every group would be killed .The same to the other 6 rats of each group 9 weeks after 5/6 nephrectomy.Urinary protein excretion and serum lipid were assayed;RT-PCR was used to examine TGF- β1 in kidney. The expression of TGF- β1 was analyzed by immuno-histochemistry.ResuIts 1)4 weeks after operation, urinary protein excretion and TGF- β1 of cordycepssinensis-treated progressive glomerularsclerosis group was substantially decreased (p<0.05) compared with the untreated 5/6 nephrectomy group.Serum total cholesterol and LDL were lower in treatment group than that in 5/6 nephrectomy without treatment (p<0.01). Immuno-histochemistry and RT-PCR revealed that the expression of TGF- β1 were also sharply lower in treatment group than that in glomerulassclerosis group.2)9 weeks after the operation,urinary protein excretion (p<0.01) ,serum cholesterol (p<0.01) and LDL (p<0.01) of cordycepssinensis-treated progressive glomerularsclerosis group.The expression of TGF- β1 (p<0.05) was reduced in the group of treatment.Conclusion Our research indicate that Cordycepssinensis can reduceprotein ,serum,cholesterin and LDL. The expression of TGF- β1 was significantly down—regulation with Cordycepssinensis intervented, and it was probably one of the mechanism of that Cordycepssinensis could prevent the progress of glomerulosclerosis. |
