Objective To investigate the effect of Cordycepssinensis on the expression ofalbumin and insulin-like growth factor-â… in rat's liver, and to explore its therapeuticeffect and mechanism on hypoalbuminemia in progressive glomerulosclerosis rats.Methods Glomerulosclerosis model was made by 5/6 nephrectomy. Thirty sixhealthy Wistar rats were equally divided into three groups at random: normal controlgroup, model group and Cordycepssinensis group. The last two groups were put 5/6nephrectomy in force, while the normal control group was just actualized sham operation.The Cordycepssinensis group was treated by Cordycepssinensis three days after 5/6nephrectomy, while the normal control group and model group were treated bycorresponded tap water. 4 weeks and 9 weeks after operation, half rats of each group werekilled. Twenty four-hour urine volume was collected,and serum albumin,blood ureanitrogen,serum creatinine and protein amount of 24 hours urine were measured.Simultaneously, ALB and IGF-â… mRNA were measured by RT-PCR in liver of rats, andthe relationship between them was examined.Results (1)Compared with the normal control group, 4 weeks after operation,theurinary protein excretion was increased(P<0.01),serum albumin was decreased (P<0.01),and expression levels of ALB and IGF-â… mRNA were all increased (P<0.05) in modelgroup. Nine weeks after operation, the changes above were more obvious, and levels ofBUN and Cr were increased (P<0.01), too. (2)Compared with model group, 4 weeks and9 weeks after operation, the urinary protein excretion was decreased(P<0.01),serumalbumin was increased (P<0.01), and expression levels of ALB and IGF-â… mRNAwereall increased (P<0.05) in Cordycepssinensis group. (3)The expression level of ALBmRNA and IGF-â… mRNA were in obvious positive correlation(r=0.932,P<0.001).Conclusion Cordycepssinensis can increase serum albumin and enhance ALB and IGF-â… gene expression in liver of progressive glomerulosclerosis rat. This might be themechanism that Cordycepssinensis can improve the hypoalbuminemia in progressiveglomerulosclerosis rats.
