Objectives Screen serum protein and paraffin-embedded tissue sections protein expression maps in patients with Diffuse large B-cell lymphoma using surface enhanced laser desorption/ionization time of flight mass spectrometry (SELDI-TOF-MS). Reactive hyperplasia of lymph node and normal human serum as control. Trying to find the protein biomarkers in DLBCL for diagnosis.Methods Using immunohistochemical method to investigate the expression of CD3, CD20, CD23, CD30,CD38, CD68, CD56, CD79α,, MCK,ALK-1和bcl-6等in the lymphoma tissues of 56 DLBCL, which were classified according to WHO classification Tumours of Haematopoietic and Lymphoid Tissues. SELDI Proteinchip Technique (WCX2 proteinchip) was used to detect the 56 paraffin-embedded DLBCLs. 16 cases normal human serum,4 cases reactive hyperplasia of lymph node serum and 7 Reactive hyperplasia of lymph node paraffin-embedded tissue as control. The data was analyzed by Biomarker Wizard software, using BPS to set up diagnostic cast.Results In the m/z range of 2-20KDa, 187 differently expressed protein peaks were detected in the 43 serum and 40 paraffin-embedded tissue as the peak's S/N>5. There are 21 significantly different protein between normal human serum and DLBCL. There are no statistical significance among Reactive Hyperplasia of lymph node serum,normal serum and DLBCL (p<0.01). There are 6 different protein between paraffin-embedded DLBCL tissue and reactive hyperplasia of lymph node tissue (p<0.01).Conclusions The specific biomarkers of DLBCL could be detected using immunohistochemical method and SELDI proteinchip technology , which might be useful for the lymphoma diagnose and prognosis.
