Abstract/Summary:
Background:Salivary adenoid cystic carcinoma (SACC) is one of the most common malignancies of the salivary glands. The clinic character of SACC is that it arises distant metastasis easy, the biggish tumor body and the adenoid local metastasis are arises distant metastasis more easy, the metastasis tumor of lung stands first on the list. The other important biological behavior is that SACC often grew along the nerve with infiltration, it brings difficulty on therapy SACC in clinic. With the development of immunohistochemical and molecule biology techniques, people have known metastasis mechanism of SACC in-depth, but the study on SACC mechanism molecule biology level is not enough relative to rest malignant tumors. The study on SACC metastasis and nerve invasion is a fat lot at home especially.Objective:The aim of this present study is to investigate the expression of NCAM and p27in SACC, to discuss the relation between clinic biological behavior and NCAM and p27 in SACC, and cure SACC availably by molecule biology consequently.Methods:Specimens from 46 cases of SACC and 20 cases of normal salivary gland (NSG) were re-examined histopathologically. The positive expression ratio of p27 and NCAM and p27 mRNA and NCAM mRNA was observed by immunohistochemical assay and interphase situ hybridization(ISH). Statistical analysis was performed with SPSS 10.0 software. Statistically significant level was considered at a=0.05, while very significant level was considered at a=0.01.Results:1. The positive rate of p27 expression is 100% in NSG and 65.22% in SACC. The high and low positive expression rate of p27 proteins in SACC showed 53.34% and 46.67% respectively. There was significant difference in the NSG and SACC2. The positive rate of p27mRNA expression is 100% in NSG and 86.96% in SACC. The high and low positive expression rate of p27mRNA proteins in SACC showed 70% and 30% respectively. There was no significant difference in the NSG and SACC (/>>0.05).3. The positive rate of p27 proteins expression is 40% in recrudescence and metastasis of SACC and 84.62% in no relapse and metastases of SACC. There was significant difference in SACC (/*<0.05). The positive rate of p27 mRNA expression is 88.46% in recrudescence and metastasis of SACC and 85.00% in no relapse and metastases of SACC. There was no significant difference in SACC (P>0.01).4. The positive rate of NCAM and NCAM mRNA expression is 65.22% and73.91% in SACC respectively, there was significant difference in the NSG and SACC (P<0.01).5. The positive rate of NCAM proteins expression is 90% in recrudescence and metastasis of SACC and 65.38% in no relapse and metastases of SACC. The positive rate of NCAM mRNA expression is 90% in recrudescence and metastasis of SACC and 61.54% in no relapse and metastases of SACC. There was significant difference in SACC and NSG, as well as in neurotropism of SACC and no neurotropism of SACC (PO.05).6. The expression of p27 proteins was a very significant positive correlation with p27mRNA(r=0.9515, PO.05) in NSG There was a very significant negative correlation between p27 proteins and p27mRNA in recrudescence and metastasis of SACC(r=-0.9707, P<0.05). There was a very significant positive correlation between NCAM proteins and NCAM mRNA in NSG(r=0.9921, PO.01) and in SACC(r=0.9157, PO.05), as well as in recrudescence and metastasis of SACC(r=0.9226, P<0.05).Conclusion:1. There is a osculation correlation between p27 proteins expression and the occurrence and development of SACC, but no correlation between p27 mRNA expression and the occurrence of SACC.2. The relation between p27 gene and tumor is in the person of p27 proteins alterations mostly, not driven largely by genetic instability.3. There is a definite correlation between NCAM proteins expression and the occurrence of SACC, as well as between mRNA expression and the occurrence of SACC. There is a osculation correlation with metastasis of SACC. The expression of NCAM proteins and mRNA might mediate in neural invasion of SACC.4. The expression of p27 was a very significant positive correlation withNCAM and mRNA in NSG There was a very significant negative correlation between p27 proteins and p27mRNA in recrudescence and metastasis of SACC. There was a very significant positive correlation between NCAM proteins and NCAM mRNA in recrudescence and metastasis of SACC.
