ObjectiveTo investigate the expression of Growth arrest and DNA damage-inducible45a(α)m RNA in normal,benign,and malignant tissues of salivary gland and its pathological significance.MethodsTotal RNA extracted from tissue samples,and then the first chain cDNA synthesis;30 cases of salivary gland tumors from human and the same number of salivary gland tissues adjacent to the tumors were used in this study. The changes of Gadd45 a mRNA expression level were detected by reverse transcription-polymerase chain reaction(RT-PCR) and Real-time PCR.Results1.Compared to adjacent salivary gland tissues,the Gadd45 a mRNA expression level in malignant salivary gland tumors was 0.214 times lower,the Gadd45 a mRNA expression level reach the level of statistical significancein.2.Compared to adjacent salivary gland tissues,the Gadd45 a mRNA expression level in pleomorphic adenoma was 0.457 times lower and in basal cell adenoma was 0.425 lower, the Gadd45 a mRNA expression level reach the level of statistical significancein.3.Compared to adjacent salivary gland tissues, the Gadd45 a mRNA expression level in warthin tumors was 1.174 higher,the Gadd45 a mRNA expression level did not reach the level of statistical significance.4.The Gadd45 a mRNA expression were lower in malignant salivary gland tumors than that in benign(pleomorphic adenoma and basal cell adenoma)salivary gland tumors(p<0.05).Conclusions(1) Real-time RT-PCR has specificity and a high sensitivity to detection for the(2) Gadd45 a mRNA expression level in salivary gland tumors;and it is reliable and easily performed.(3) The Gadd45 a mRNA expression level in benign and malignant salivary gland tumors were decreased compared to adjacent salivary gland tissues.The Gadd45 a mRNA expression were lower in malignant salivary gland tumors than that in benign(pleomorphic adenoma and basal cell adenoma)salivary gland tumors.The results suggest that the Gadd45 a may play an important role in in the tumorigenesis of salivary gland tumors by inducing the tumors continue to proliferation.(4) Gadd45 a might be used as a potential molecular target for the treatment of salivary gland tumors.
