Objective To explore the functional mechanism ofphospholipase A2 in the low back and leg pain caused by herniated lumbar discs.Methods (1) The general clinical materials –history of disease,age,sex wascollected and the pain grades of patients of herniated lumbar discs (herniated groups)and controlled groups was evaluated with Oswestry disability Index one day beforeoperation; (2) the pulposus of the herniated and control groups was collected inoperation and divided into two parts, with one of them stored at -70℃. The secretoryand cytosolic phospholipase A2 were assayed in the frozen sections byimmunostaining with elivision method; (3) another samples was homogenized andcollected the supernatant for biochemical assay after centrifuging; (4)The statisticalanalysis with SPSS 11.5 software was performed.Results (1) The biochemical activities and immunochemical expressed rates ofsecretory and cytosolic phospholipase A2 in the pulposus of the herniated groups werehigher the control groups, and were correlation with the grades of pain. (2) Thesecretory and cytosolic phospholipase A2 were mostly located in the disc cell,expecally in poly-nuclei disc cells. ( 3 ) The biochemical activities andimmunochemical expressed rates of secretory and cytosolic phospholipase A2 in thepulposus of the wereConclusion The phospholipase A2 may play important pathomechanisc in the lowback and leg pain caused by herniated lumbar discs, and there may be some correltionbetween the phospholipase A2 and the degenerated pulposus.
