Mechanism Of Matrine On Apoptosis Of Rat Hepatic Stellate Cell In Vitro

Objective To investigate the mechanism of matrine on apoptosis of ratHepatic Stellate Cell in vitro.Methods HSC line rHSC-99 was incubated with matrine of differentconcentration of 0.05mg/ml ,0.1mg/ml ,0.2mg/ml ,0.4mg/ml for24h,.RT-PCR was used to detected mRNA of p75 and fas of HSC. Theearly stage apoptosis was detected by Annexin v-FITC/PI double labbledassay.morphology of apoptosis was detected by transmission electronmicroscopy. Results Administration of 0.05mg/ml-0.4mg/ml matrinecould significantly induce HSC apoptosis in dose-dependent of manner,Expression of P75 of different concentration matrine treated HSC wereenhanced in dose-dependent of manner,but no significant change in fas.Electron microscopy showed that nucleoli disappeared,chromatincondensted,ranked along inside of nucleonic membrane. conclusionmatrine can increase p75 expression of HSC which may be related to itsinducing apoptosis of HSC...
