Objective:We add different concentration of matrine to human fetal hepatocytes culture fluid DMEM,then observe and record fetal hepatocytes growth,synthesis fuction,secretion fuction,apoptosis and the expression of XIAP to find the better hepatocytes culture fluid in which fetal hepatocytes can maintain activity and grow quickly,in order to establish platform for biological reactor,hepatocyte library and hepatocyte transplantation.Method:We harvest human fetal hepatocytes from fetal human liver,then culture fetal hepatocytes in DMEM added 2mg/L,8mg/L,16mg/L,32mg/L and 64mg/L matrine,and detect ALB,ALT,UA,apoptosis by FCM and the expression of XIAP by RT-PCR at 12h,18h,24h,48h and72h.Rsult:Except for 64mg/L matrine group,the human fetal hepatocytes cultured in different concentration of matrine showed different morphological manifestation,ALB,ALT,UA,apoptosis ratio and expression of XIAP from control group.In 2mg/L-64mg/L matrine groups,the level of ALB,UA and expression of XIAP is higher than control group,ALT and apoptosis ratio is lower than control group.Conclusion:Matrine can protect fetal hepatocytes from injury and maintain the activity,it can also inhabit the apoptosis of fetal hepatocytes,and 8mg/L is the best concentration.Up-regulation of XIAP expression may be the protection mechanism on molecule level. |
