matrine induces apoptosis in S180-saroma in micespecialization integration of Basic traditional Chinese and Westernmedicine MS candidate Shi yan advisor Liu chun ying professorObjective: To investigate the effects of matrine on the cellgrowth and apoptosis in S180 sarcoma in mice.Method: The model for transplanted S180 fluid tumor in mice wasset up. 40 Balb/c mouse, 18-22g, were separated to 4 group randomlyaccording to mass (n=10 each ), S180 sarcoma cell were implanted intoall groups by subcutaneous injection. After 48h, the 4 groups micewere given 0.9ï¼…NaCl 0.2mL, matrine (50mg/kg), CTX (20mg/kg), matrine(50mg/kg)+CTX (20mg/kg). After the drug was given continuouslyfor 15 days, the mice were killed by decapitation, caculate tumorinhibiting rates by tumor weight .Observe matrine' s inhibition ofthe S180 sarcoma in mice from developing by pathological diagnosis invivo ascitic; observed the expression of Bcl-2 protein by SABCimmunohistochemistry method and apoptosis by electron microscope.Result: 1. General state of the 4 group:matrine had more smoothfur and better psychosis ,moved less targily than CTX; matrine+ CTXhad smooth fur, good psychosis, more food, moved agiliy. 2. Each groupwith with drug had effect on inhibiting S180 sarcoma cell, LNT+DDP hadthe best effect, 43.6ï¼…, the rate of inhibitory effect of LNT, DDP was32.4ï¼…, 35.5ï¼…respectively. 3. Under optical microscope, the cell of NSarrayed effuse, the nuclear was big ,deep-stained, thickened-karothe-ca, chroma, granul-ated, Pathologic, karyokinesis, can, be, seen. Proliferation of fibrous tissue ,necrosis and infiltration can be seenin CTX group; There were little apoptosis ,karyopycnosis ,condensed-plasm, deep-stained ,and no infiltration in group MATRINE ; Moreapoptosis in MATRINE+CTX;Under electron microscope the cell in NSwas round or irregular, there were a lot of micro-villus on the cellsurface, nuclear membrane was irregular with angular bites, manynucleolus, no swollen mitochondrion;the cell became smaller,mitochondrion was swollen, the chromatin crescent shaped along theinner margins of the nuclear; There was chromatinorrhexis in groupCTX; There was karyopycnosis, chromatin condensation along the innermargins of the nuclear, more lysosomes could be found in cytosol,the mitochondrion became bigger and rounder, the mitochondrialcrista became less and shorter, even disappeared. In condition of very swollen, it could became the structure of vacuole, and apoptosisbody, which is the characteristic structure of apoptosis, could befound. 4.0ncogene Bcl-2 protein was stained in cytoplasm, Bcl-2protein expression in matrine, CTX, matrine+ CTX reduced after givendrug, the effect of matrine+ CTX is best.Conclusion: 1. Matrine can raise the life quality of S180 sarcomamice. 2. Matrine can inhibit the tumor of S180 sarcoma mice. 3. Matrinecan induce apoptosis of S180 sarcoma cell. 4. Matrine can regulate theapoptosis-related gene Bcl-2 expression. 5. Through inducing apopoto-sis, regulate the Bcl-2 expression, Matrine raise the life quartilyAnd inhibit the tumor growth. 6. Matrine has co-operated with cytoxan.
