Background and Objective: Tissue microarray is a special biologic microarray. which has the behavior of small volume, high throughput (information), and design by different demand. It is suitable to immunohistochernistry, DNA or RNA in situhybridization with high efficiency and flexibility, and its results are highly uniform with remarkable credibility and comparability. Compared with the traditional pathological investigation: tissue microarray is an efficient. convenient and standardized method applied to research in oncology. This current study was to establish the tissue microarray technique of manual method for the preparation of tissue microarray and to explore their application in immunohistoche mical stains.Survivin is a recently idenfied member of the inhibitor of apoptosis protein (IAP )family ,which inhibits processing of down streamcffect or caspase 3 and caspase 7 in cell receiving a poptotic stimuius.Unlike other IAP protein , found during embryonic and fetaldevelopment. Survivin was completely down regulated and undetectable in normal adult tissues, and became prominently reexpressed in all of the most commoncancers. Survivin plays important pole in regulation of cellcyclc. Survivin targeting antisense nucleotide and surviving gmutants in ducc apoptosis. this current study was designed to investigate the effects of survivinin tumorigenesis and progression of breast carcinoma though observing the expression of survivin in the tissue of normal mammary and breast carcinoma.Materials and Methods:A total of 82 1 .Omm samples from 82 paraffin embedded blocks of mazoplasia and breast cancer were constructed in this study .Routine 4pm sections were cut from the TMA blocks . Haematoxylin-eosin immunohistochemical stains were performed on the sections .The expression of xurvivin in the tissue of normal mammary and breast cancer by imrnunohistochemistry.The relationship between survivin expression and the pathological features of breast cancer was assecssed.Resualts:1 . The dots on TMAs were regularly arranged and their morphology were well preserved.Hamatoxylin-eosin staining showed that 100% samples in paraffin section of TMA could be analyzed morphologically.Expression of survivin, p53, bcl-2 were detected in the most dots of TMA sections. Well-preserved morphology and immunohistochemical activaties were also observed in at 100%dots of the TMA.2. The expression of survivin in the tissue of normal mammary was negative.The positive ratesofsurvivinwere23.81%(5/21), 70.00% (7/10), 75.00% (15/20), 77.42% (24/31) in the tissues of mazoplasia, ductal carcinoma in situ, infiltrative ductal carcinoma without lymph node metastasis, infiltrative ductal carcinoma with lymph node metastasis. In the last three groups were higher than those in the first groups ^<0.05).The overexpression of survivin was correlated with the pathology grade of breast intraduetal carcinomas although the difference is not significant (P>0.05).3. The positive rates of bcl-2 were 38. 10% (8/21), 50.00% (5/10), 65.00% (13/20) #1 67. 74% (21/31) in the tissues of mazoplasia, ductal carcinoma in situ, infiltrative ductal carcinoma without lymph node metastasis, infiltrative ductal carcinoma with lymph node metastasis. In the last three groups were higher than those in the first groups (7><0.05).The overpression of bcl-2 was correlated with the pathology grade of breast intraduetal carcinomas although the difference is not significant (P>0.05).4. The positive rates of p53 were 33.33%(7/21 ),40.00%(4/10), 65.00%( 13/20), 87.10% (27/31) in the tissues of mazoplasia, ductal carcinoma in situ, infiltrative ductal carcinomawithout lymph node metastasis^ infiltrative ductal carcinoma with lymph node metastasis. In the last three groups were higher than those in the first groups (P<0.05).Conclusions:1. This current study was to establish the tissua microarray technique of manual method .well-preserved Heand immunohistochemical activaties were observed in ai 100% dots of the TMA.it is feasible to utilize tissue microarray for a rapid > economic and accurate screening.2. It have the same result that the expression of surviviv with tissue microarray and immunohistochemical stains were used to detect the expression olsurviviru bcl-2, p53,compared with the result with the traditional pathological investigation.3. The expression of survivin in the tissue of normal breast was negative.The overexpression of survivin in breast cancer, expression of survivin was correlated with. the pathology grade of breast intraductal carcinomas.4. The expression of survivin in breast cancer higher than the tissue of mazoplasia .Hxpression of survivin was correlated with ductal carcinoma in situ. infiltrative ductal carcinoma without lymph node metastasis. infiltrative ductal carcinoma with lymph node metastasis although the difference is not significant.5. Survivin may play an important role in the development and progression of breast cancer.There was a positive correlation of survivin expression with bcl-2 and p53.
