Abstract/Summary:
objective: This study investigates the damage of myocardial cells induced by adriamycin or X-ray alone in cultured rat cardiac cells. and investigates the damage of cardiac muscle cells after combined adriamycin and X-ray.And investigates the possible damage mechanism of combined adriamycin and X-ray induced myocardial damage. Methods: After exposed to adriamycin or radiation single or combined adriamycin and X-ray,viability of cardiac muscle cells assessed by MTT colorimetric assay, Myocardial of lactic acid dehydrogenase (LDH) was examined by blood biochemical autoanalyzer.Meanwhile,the Morphological characters of apoptosis were checked with laser-scanning confocal microscope by AO/PI staining and cardiac myocytes apoptosis rates were detected by flow cytometry. Results: (1) adriamycin and X-ray inhibited the growth of rat cardiac muscle cells in the dose-dependment or time-dependment manner in a certain range of dose. And myocardial of lactic acid dehydrogenase (LDH) revealed were increased by adriamycin and X-ray single; (2) the Morphological characters of apoptosis induced by adriamycin(5.00μg/ml) and X-ray(40Gy) were surveyed by laser-scanning confocal microscope in cultured rat cardiac cells;(3) adriamycin and X-ray induced apoptosis rate detected were increased by flow cytometry;(4) In the model, combined adriamycin(5.00μg/ml) and X-ray (40Gy) obviously inhibited cell viability of cadiocytes compared to both adriamycin and X-ray alone by MTT assay. The consistent of lactic acid dehydrogenase (LDH) and apoptosis rate was augmented more obviously in myocardial cells. Conclusions: Apoptosis may be involved in the cells damage caused by exposure of cultured rat cardiac muscle cells to Adriamycin or X-ray. The cells damage were more aggravation by exposure of cardiac muscle cells to combined adriamycin and X-ray. The mechanisms of this damage were possible different, Adriamycin and X-ray probably played different role in cadiocytes apoptosis via diverse mechanisms.
