Neurasthenia is a common neurosis,which has high excitability and fatigabilityof mind with worried mood and physiologic and pshychological disorder, itusually happens to those who have persistent mental labor andover-exertion fatigability and insufficient sleep. The main symptoms areinsomnia headche,dizzy and memory descent, sometimes it could have othersymptoms, such as circulation , digestion, endocrine , metabolism andgenital system .Patients often feel uncomfortable with the whole body , butthere is no abnormal findings in body check . the course of disease would lastfor a long time and could effect their health and work.At present ,modern medicine are failure to treat it efficiently . Mosttranquilizer can be used to relieve tension and improve somnus , but themalso could cause some side effects such as dizzy, chrousness and dugresistance.There, it has important theropy meaning to seek safe and activemethods to treat neurasthenia.According to Chinese traditional medical theory, Naolingsu capsule isprodused ,it contains huangjing, ginseng, antler pilous,Chinese magnoliavinefruit, colla cornus cervi,polygala, jujube seed, fructus, tortoiseshell,ophiopogonis,hoelen,Siberian cocklebur fruit,Chinese jujube,epimedium herbetal.Some of these compositions have the function that tonifying energy andblood reinforcing hepar kidney, increasing overcast,strongthen Yang,tranquizing mood and calming tension,fasten bones and muscles. So it couldbe used to treat neurasthenia , insomnia, poor memory, dizzy,cardiopalmus,fatigue,hypodynamia,body deficiency,perse sweat asynodia emissions et al.The research study the pharmacodynamics of naolingsu capsule, whichinclude the effect on independent activity of mice;the effect on hypnosis ofpentobarbital sodium of subthreshold dose; the effect on hypnosis ofpentobarbital sodium; the effect on antieclampsia; the ability ofanoxia-resistance of mice; the ability of fatigation-resistance of mice, theability of cold-resistance of mice; the effects on yang asthenia mice;theeffects of cold-resistance and fatigation-resistance of splenic astheniamice;the effect on reticulo-endothelium phagocytosing function hypnosis ofnormal mice and the effects of Naolingsu capsule on peripheral bloodleucocytes(PBL) and immune organs'weight of hypoimmunity mice. Andprovide experimental basis for clinical application.1.MehtodsWe gave mice Naolingsu Capsule of different dose by gavage, then weobserved the following indexes 1h later:1.1 The effect on independent activity of mice: comparing the independentmotion times( in 10 minutes) before and after administration.1.2 The effect on hypnosis of pentobarbital sodium of subthreshold dose:giving mice the subthreshold dose pentobarbital sodium, then observing thesleep occurrence percentage.1.3 The effect on hypnosis of pentobarbital sodium : giving mice thesubthreshold dose pentobarbital sodium ,then observing the disappearing timeof righting reflect and sleeping-time of mice.1.4 The effect on antieclampsia: making mice to appear tonic convulsion withthe certain electric voltage, current intensity, then observing the death rate andtonic rate in the 0.25 and 11.00 second.1.5 The ability of anoxia-resistance of mice: putting mice into an airtight jar of125 ml, in which there is dry sodalime of 5 g, then observing the survival timeof mice.1.6 The ability of fatigation-resistance of mice: Mice were placed in a bathingcontainer, with 30-cm-height and 27±1℃water ,then the swimming durationwas recorded.1.7 The ability of cold-resistance of mice: Mice were placed in an ice cabinetwith temperature-15±1℃, then the survival duration was noted..1.8 The effects on yang asthenia mice: 1h after hydrocortisone was given byintramuscular, the weight, body temperature and low temperature swimmingsurvival duration of mice were recorded.1.9 The effects on cold-resistance and fatigation-resistance of splenic astheniamice: Mice were placed in an ice cabinet with temperature-15±1℃, thenthe survival duration was noted. Mice were placed in a bathing container, with30-cm-height and 27±1℃water, then the swimming duration was recorded.1.10 The effect on reticulo-endothelium phagocytosing funtion of normal mice:The India ink was injected into vena caudalis of mice , with dose of 10 mls/ kg,1 min and the 5 min later, 20 ul venous blood was drown from vena orbitalis,and then was dissolved into 2 ml 0.1% Na2CO3 evenly, the optical densitywas measured .At last, we calculated the index of phagocytosis k, weighed theliver and spleen, and then got the activity of phagocytosis α.Furthermore,we also observed the effects of Naolingsu capsule on peripheralblood leucocytes (PBL) and immune organs'weight of hypoimmunity mice:In the 2,4,7 day after administration, we drew the venous blood from tail veinand then counted PBL .Then we killed mice, and weighed thymus glands andspleens.2 Results2.1 The effect on independent activity of miceIn the 2,4g/kg of Naolingsu capsule group, the times of autonomic movementof normal mice have obviously difference contrast to the controlone.(p<0.05,p<0.01),which the 1g/kg of Naolingsu group have the oppositeresults.2.2 The effect on hypnosis of pentobarbital sodium of subthreshold doseIn the 2,4g/kg of Naolingsu capsule group,the perconbuge of slpeepinghappening obviously higher than the control one (p<0.05 or p<0.01),which the1g/kg of Naolingsu capsule have difference between the testing group and thecontrol one(p<0.05),which suggests the Naolingsu capsule can coordinate thehypnosis of pentobarbital sodium under bubthreshold dose.2.3 The effect on hypnosis of pentobarbital sodium.We can find in the 2,4g/kg of Naolingsu capsule group, the time of fallingasleep and sleeping, have obviously difference contrast to the control one,howerer there is no difference in 1g/kg's group.2.4 The effect on antieclampsiaIn the 1,2,4g/kg of Naolingsu capsule group, the rate of decrease stiffness anddeath have no diffdrence contrast to the control one, which suggests thiscapsule has no the role of anti-convulsion.2.5 The ability of anoxia-resistance of miceIn the 1,2,4g/kg of Naolingsu capsule group,the capsule can extend thesurvival time contrast to the control group.2.6 The ability of fatigation-resistance of miceIn the 2,4g/kg's group the capsule can extend the swimming time with theload of mice and resist fatigue .In 1g/kg's group it can extend swimming timeas well ,but there is no diference.contrast to the control one.2.7 The ability of cold-resistance of miceThe capsule can enhance the abilities of cold-resistance and low temperatureswimming survival duration in all group ,but there is no difference in the1g/kg's group,otherwise the 2,4g/kg's group have the opposit results.2.8 The effects on yang asthenia miceIn the control group ,the normal mice put on weight by 1.9g in three days atthe beggning of the test,while the yang asthenia group lost their weight by1.8g .In the 1, 2,4g/kg's group ,the capsule can delay the decline of weightcaused by hydrocortisone ,which have no difference contrast to the yangasthenia group(p>.0.05).In the control group ,the average body temperature is36.2±0.6℃,but in the yang asthenia group the average body temperature is35.7±0.4℃,the obviousl deference suggest that Naolingsu capsule remainthe temperature of the animals of the yang asthenia . Contrast to the normalmice, the yang asthenia mice get a descent ablility of cold-resistence andfatigution-resistance.the low temperature swimming survival duration in the2,4g/kg's group can be seen longer, but in 1g/kg's group is no obvious.Change.2.9 The effects on cold-resistance and fatigation-resistance of splenic astheniamiceIn the sptenic asthenia group ,the ability of cold-resistance andfatigation-resistence decline contrast to the normal mice .the Naolingsucapsule can extent the low temperature swimming survival duration andadvance the ability cold-resistence in the 2,4g/kg's of Naolingsu group.In1g/kg of Naolingsu group capsule also can extent the time , but there is nodifference contrast to the control group .In 2,4g/kg'sgroup , the capsule allcan obviously extent mice's swimming time under load.. we also find that thecapsule can resist fatigue .In the 1g/kg of Naolingsu group ,the capsule alsocan play the same role,while no obvious difference to the control group.2.10 the effects of Naolingsu capsule on peripheral blood leucocytes(PLB)and immune organs'weight of hypoimmunity miceCyclophosphamide can decline the weight of the thymus gland and spleenand the peripheral blood leucocytes. In the 2,4g/kg'sgroup, Naolingsucapsule can obviously resist the decrease of PLB and the decline of weight ofthymus gland and spleen, which caused by cyclophosphamide.but the group of1g/kg Naolingsu is not obvious.2.11 The effect on reticulo-endothelium phagocytosing funtion of normalmice:In 2,4g/kg's Naolingsu capsule group , the k and a is higher than the controlgroup , which suggest the capsule can enhance the reticulo-endotheliumphagocytosing funtion of normal mice..3 Conclusions...
