Objective To investigate the effects of ShenFu Injection on myocardial injury induced by Isoprenaline, and to explore the underlying mechanisms. Methods To establish the myocardial injury model induced by Isoprenaline (Iso), Isoprenaline was administered (30mg·kg-1·d-1) to Sprage-Dawley rats once a day ,for two days. ShenFu Injection was administered in combination with Isoprenaline in order to study the ShenFu Injection' effects on Iso-Induced myocardial injury. We measured cardiac index, CK, LDH content in serum, myocardial pathological changes, Na+-K+-ATPase activity, superoxide dismutase (SOD) activity,glutathione peroxidase (GSH-PX) activity , the cardiac malondialdehyde (MDA) content and the expression of bcl-2, bax in cardiomycyte. Results In the Iso group, cardiac index was increased, compared with thecontrol group, and CK> LDH content in serum were higher than control group, the same to cardiac malondialdehyde (MDA) content. The activities of Na+-K+-ATPase ^ GSH-PX and SOD are all decreased in the Iso group .Cardial injury was found in the Iso group through pathological examination. Judging from the pathological examination, ShenFu Injection made cardial injury induced by Isoprenaline worse. ShenFu Injection can also promote the increase of CK, LDH content in serum, induced by Isoprenaline. Both the activities of GSH-PX and SOD in the group of ShenFu Injection in combination with Isoprenaline were higher than those in the Iso group. On the contrary, ShenFu Injection can accelerate the decrease of Na+-K+-ATPase activity induced by Isoprenaline, dose-dependently. There weren't differences among the groups of the ISO and ShenFu Injection in combination with Isoprenaline. In the Iso group, the expression of bax in the cardiomycytes was increased and the rate of bcl-2/bax was decreased. After the combination of the ShenFu Injection at the biggest dose in this experiment, the increase of the bax expression induced by Isoprenaline was accelerated and the rate of bcl-2/bax was also decreased further, but not at the other two doses. Neither the Isoprenaline nor the ShenFu Injection affected the expression of bcl-2 in this experiment.Conclusions ShenFu Injection can strengthen the myocardial injury induced by Isoprenaline (30mg ? kg"1 ? d"1). The mechanism of ShenFu Injection on itsstrengthening on myocardial injury induced by Isoprenaline refers to its acceleration on the decrease of Na+-K+-ATPase activity and the increase of the expression of bax which both can be caused by Isoprenaline.
