Study Of Serum Levels Of IL-8,IL-12, TNF-α And IFN-γ In Patients With Condyloma Acuminatum

Condyloma acuminatum(CA) is a hyperplastic disease caused by Human Papillomavirus(HPV) and locates at external genitals and perianal area. The morbidity of CA has increased in recent years. The disappearance, persistence and recurrence of CA are related to cellular immunity function disorder. Cytokines play a leading role in cellular immunity. The unbalance of Th1 and Th2 cellular function in-patients can induce Th1 cytokine expression down-regulation and Th2 cytokine up-regulation. This phenomenon can result in change of the level of cytokine and virus can not be removed effectively. We presume that CA patients exist abnormality of cytokines expression.In order to study the cellular immunity state of CA patients, we used double antibody sandeich ELISA for quantitative determination of serum IL-8, IL-12, TNF-α and IFN-γ levels in 40 patients with CA. Our results showed:1. The serum levels of IL-8, IL-12, IFN-γ were significantly lower inCA patients group than that of controls group, but The serum level of TNF-a was significantly higher in CA patients group than that of controls group. This outcome was consistent with internal and external reports. It indicated that CA patients existed cellular immunity disorder especially decreased expression of Thl cytokines.2. Serum level IL-8, IL-12, TNF- a and IFN- Y had no significant differences between CA first-outbreak group and recurrence group, course of disease >3 months group and course <3 months, course of disease >6 months group and course <6 months. These results were not consistent with literatures. This was likely to be caused by few cases.3. By analysing the correlation among these cytokines of CA patients, we found that among the serum levels of IL-8 and IL-12, IL-8 and IFN-Y ,IL-12 and TNF- a JL-12 and IFN- Y ,TNF- a and IFN- Y had significantly positive correlation, but there was no correlation between the serum level of IL-8 and TNF- a . We found that there were definite correlativity among the cytokines of CA patients, but the reason is unknown at present and more investigations are needed.To sum up, the human body can appear cellular immunity disorder after being infected with HPV. The functions of Thl and Th2 in CA patients are in unbalance. All of the Thl cytokines reduced in CA, otherwise, Th2 cytokines relatively increased. Therefore, the expression ability of Thl cytokines such as IL-8, IL-12, TNF-a and IFN-y changed and the ability of cleanning virus reduced then lead to coming on CA.. In clinic we can use ELISA method to measure the serum level of cytokines in CA patients.to help us understand the dynamic change of cytokines in CA, and at the same time itcan supply important laboratory evidence for treating CA patients with cytokines.lt also helpful to prevent and treat CA.
