Objective1. To set up the immuno-histochemical method for testing HPV6-E7 protein.2. To analysis the expression of HPV6-E7 protein in normal cervix uteri tissues, incipient and replase condyloma acuminatum tissues.3. To explore the relation between HPV6-E7 protein and replase of condyloma acuminatum, age and sex of patient.4. To investigate the biological significance of HPV6-E7 in conversion of condyloma acuminatum.Methods1. HPV6-DNA of patients was determined using fluorescent probe PCR.2. HPV6-E7 was determined using immunohistochemical staining in normal cervix uteri tissues, incipient and replase condyloma acuminatum tissues. PBS was used as HPV6-E7 negative control.3. Cellular localization: HPV6-E7 staining reaction was seen in the nuclear. Immunostaining was evaluated as follows: ~, none of neoplastic cells with positivity; +, ≥15% of neoplastic cells with positivity.Results1. There were 91 cases infected HPV6 in 152 condyloma acuminatum patients. 56 cases were incipient condyloma acuminatum, 35 cases were replase condylomaacuminatum; 53 cases were 18-38 years, 26 were 39-48,12 were 49-60; 52 cases were male patients, 39 were female patients.2. The positive rates of HPV6-E7 protein.was 70.3 %(64 /91) in the tissues infected HPV6 of 91 cases. It was 0, 69.5% (39/56) and 71. 4% (25/35) in normal cervix uteri, incipient and replase condyloma acuminatum respectively. The positive rates of HPV6-E7 in incipient and replase condyloma acuminatum tissues were significantly higher than those in normal cervix uteri(P< 0.001). There is no difference in statistics in incipient and replase condyloma acuminatum tissues(P>0. 05).3. The positive rates of HPV6-E7 protein in patients of 18-38 years (53cases^ 39-48years(26cases)and49-60years(12cases) were71.7% > 69.2% and 66.7 % respectively. They have no difference in statistics.4. The positive rates of HPV6-E7 protein in male patients (52 cases) and female patients(39 cases) were75 % and 64.1 % respectively. They have no difference in statistics too.ConclusionHuman papillomavirus type 6 (HPV6 ) is one of the most important pathogenic element of condyloma acuminatum in our country(The infection rate was 59.9%). There was higher positive rates of HPV6-E7 protein in the tissues infected by HPV6 of 91 cases(70.4%). The positive rates of HPV6-E7 protein in incipient and replase condyloma acuminatum tissues were significantly higher than those in normal cervix uteri, and has no significant difference in incipient and replase tissues, there is indifference in age and sex too. Our study results suggest that HPV6-E7 protein is correlative with the invasion of condyloma acuminatum, and it also plays an important role in the development of condyloma acuminatum. Though there is some relation with its replase, they have no statistical significant. It needs further study for the relation between HPV6-E7 protein and the conversion of condyloma acuminatum. There is important significance to study thorough HPV oncoprotein especially HPV6-E7 protein in prevention and cure of condyloma acuminatum and study ofeffective vaccine. |
