[Objective]To investigate the abnormal expression of Fas, C-erbB-2, PCNA , P16 and relationship of HBV and HCC by detect the expression of Fas, C-erbB-2, PCNA and P16 in chronic hepatitis B, and to study the expression and value of Various type of genes, and provided theoretical foundation for gene therapy. [Methods]1. Immunohistochemical staining: 87 samples of chronic hepatitis B and 30 cases HCC were selected and the expression of Fas, PCNA, P16 were detected and 62 samples of chronic hepatitis B and 55 cases HCC were selected and the expression of C-erbB-2 were detected by immunohistochemical technique(LSAB).2. Cellular localization: Fas and C-erbB-2 staining reaction was seen in the cytoplasm and Cell membrane , Proliferating cell nuclear antigen(PCNA) and P16 was nuclear staining. Immunostaining was evaluated as follows: -, none of liver cells with positivity; +, ≤25% of liver cells with positivity; ++, from 26% to 75% of liver cells with positivity; +++, ≥ 76% of liver cells with strong positivity.[Results]1. The expression of Fas in chronic hepatitis B and HCC :The Faspositive-expression rate in ASC^ CPH, CAH and HCC Was 81.3%, 80. 8%> 62.2% and 33.3%, respectively.2. The expression of C-erbB-2 in chronic hepatitis B and HCC :The C-erbB-2 positive-expression rate in ASC> CPH> CAH and HCC Was 44. 4%^ 93.8%, 95.2% and 52.6%, respectively.3. The expression of PCNA in chronic hepatitis B and HCC :The PCNA positive-expression rate in ASC, CPH> CAH and HCC Was 50.0%, 57. 7°/^ 73. 3% and 96. 7%, respectively.4. The expression of P16 in chronic hepatitis B and HCC :The P16 positive-expression rate in ASC, CPH, CAH and HCC Was 37. 5%> 23. 1%, 15.6% and 3.3%, respectively.[Conclusion]1.there is a high expression of Fas in Chronic hepatitis stage and a low expression of Fas in HCC, while the positive-expression rate of C-erbB-2 was higher than that of Fas in CPH CAH, but the positive-expression rate of C-erbB-2 was lower than that of Fas in ASC, indicate that Fas and C-erbB-2 were correlate significantly with liver cell immune evasionand cancerationin in various liver damaging stage, and that apoptosis mediated by Fas and FasL may be one cause responsible to clean the liver cell infected HBV and immune evasion,2. Chronic hepatitis stage, there is a certain degree loss of P16, PCNA has a high expression, while there is a strong expression of PCNA and a marked loss or absence of P16 expression in HCC, indicate that the expression of oncogenes and cancer suppressor genes play a important role in primary hepatocellular carcinoma.
