| Licorice is processsed from dry root or rhizoma of leguminosae glycyrrhiza plant glycyrrhiza ulalensis fisch,glycyrrhiza inflata bat, or glycyrrhiza glabra L.It has the function of normalizing function of the stomach and pleen,moistening the lung,detoxifcation,unison various medical.At present,the quality evaluation indicator of crude-licorice herbal pieces prepared for decoction is single and lack of scientificaalness;the processing condition of roast-licorice is not fine and without reliable quality standard.In order to solve these actual problems,the experiment used quality licorice which from different producing area,different level and different detachable time,to strike up crude-licorice and roast-licorice 'finger-print utilizing HPLC.Established the HPLC quantitative test method of glycyrrhizicacid and glycyrrhizin at the same time;optimized the methods of roast and microwave heating,in order to substitute traditionary saute craft.1.Established the HPLC quantitative test method of glycyrrhizicacid and glycyrrhizin at the same time,both degree of separation is good,regression equation correlation coefficient run up to 0.9999 upwards.2.We selected glycyrrhizin and glycyrrhizicacid' weighting content by way of evaluating indicator,useing uniformity design method ascertained best precondition condition of finger-print,which was cold maceration 10h,ultrasonic sound distilling 30min by 50% methanol.3.Useing 17 different samples,we established the crude-licorice' HPLC finger-print.The finger-print whole degree of separation was welldefined;methodological reproducibility,stability and sample' uncommonable apex scale,retention time' RSD all conformed finger-print'requirement.4.We striked up the roast-licorice' finger-print by 10 different samples,which according to traditional craft process strictly.5.Selecting finger-print'degree of similarity for evaluating indicator and useing orthogonal design,we optimized the the best method of roast,which was crude-licorice plusing 25%refined honey(mixed 30%water),close moistened 30h,90℃roasted 4h.And the method' reproducibility excelled the method of saute' in evidence.6.Selecting finger-print' degree of similarity for evaluating indicator and useing orthogonal design,we optimized the the best method of microwave heating,which was crude-licorice plusing 25%refined honey(mixed 30%water),close moistened 30h,40%sped microwave heating 6 mins.And the method' reproducibility excelled the method of saute' in evidence,even better than the method of roast'. |
