Tumors are progressive and severe diseases in which malignant tumors are very harmful people's health. The obvious studies showed that tumor has tight relitionship with immune functions. The factors of both sides affects development and post-treatment for these type of diseases. The incidence rate of tumor would be increased when low immune response occurs in the hosts. While, the incidence rate decreased when the tumor is not under conditions of progression. So, the scientists working in the medical fields focus on promotion of immune functions to destroy the tumor cells. At the present time, some anti-tumor chemicals or combined with radiation-therapy has been researched and developed to have prolong the life span, though, undesired or side effects has been constantsly happening. All of these may have troubles for treatment of tumor coming with problems of drug's genesis. All over the world, the interests of anti-tumor drugs has changed to natural herbs gradually. Our traditional Chinese medicine and herbs are being reconsidered at all. For past 50 years, with accurated diagnosis for tumor, great progression has been made in both clinic and basic researches. However, crude extracts were applied in the ex vivo or general experiments,somewhat, it couldn't represent the actual effective substance to carry out it's job. In addition, crude extracts affect growth of cells with their physical or chemical features. Since crude extracts own some interfered factors for experimental results, we have to apply some serological approaches in traditional Chinese herbs. It could both reflect the directed interaction of soluble part of drugs and undirected interaction bwteen drug's following metabolates and intra ligands. Juglans mandshurica maxim is subcategoried species of Juglans regia. It has been used in treatment gut and bladder tumor since ancient time. Walnut injection also demonstrates therapeutical effects on tumors as well. But there is no reports in treatment of tumor in details. So, S180 cells was used to identify the functions of Juglans mandshurica maxim. The extracts were prepared from Juglans mandshurica maxim by the way of ethanol soaking, low pressure distill, organic reagent abstraction. Finally, the desired part of Juglans mandshurica maxim was got by means of chromatography. Primary experiments showed that LD50 and 95% range is 640.12-395.38mg/kg respectively. In the inhibited experiments on proliferation of cells, S180 cells were innoculate to the cavity of mice. During the logarithm growth phase, extracts from Juglans mandshurica maxim with various doses were added. 5-Fu was used to be positive controls and cultured medium used to be blanking controls. MTT method was used to detected OD values. Inhibited ratio was calculated and curve was drew. Serological pharmacological tests were carried out. Intragastric 2 times for sequential 3d. These mice were fasted diet for 12h while no water banned. The blood were taken after 1 h administration, thenwas inactived under 56 for 30 min. The serum with drug was treated by steriled methods. The various doses were given to mice by intragastric administration. With the same dose to the mice, various extracts were added to be various dosage groups. MTT method was used to detected OD values to identify the optimized diluted solution. Some immune index such as lymphocyte transformation function, NK cells activities, TNF activities and IL-2 induced level were determined Results: Within the range of 4.125-33.00ug/ml, the extract of Juglans mandshurica maxim show remarkable inhibited function on proliferation of S180 cells. From the inhibited curve, it also demonstrates that this function is dose-dependence. 5% serum with the extract of Juglans mandshurica maxim is the best among 5-20%. Various dose show inhibited activities and no noticed difference compared with positive controls. The extract of Juglans mandshurica maxim show significant activities on anti-tumor. In inhibited ratio were 32%, 43%, 56% with low, medium, high dosage respectively. In addition, mice showed no obvious side effects such as anepithymia,vomiting et. al. The extract of Juglans mandshurica maxim are suggested to increase weight of thymus and increase thymus index.
