Ischemia heart disease (IHD) is also called coronary heart disease (CHD), in the light of traditional Chinese medicine (TCM) it belongs to the scope of "angina cordis"or "cardialgia", ect. Recent years, the disease rate of CHD is rising in our country and the people's health is threatened seriously by CDH. Therefore it's of great significance to explore and develop the effective drugs which are used to prevent and treat CHD. DangguiShaoyaoSan (DSS) was originally an effective female prescription in Jingui Yaolüe. It's made up of DangGui, ShaoYao, ChuanXiong, FuLing, BaiZhu, ZeXie. Recent years it has been used to treat many diseases in every clinical field according to the principle of "strictly observing the pathogenesis and treating the different disease in the same way". My tutor has found it is beneficial to CHD and we haven't retrieved the empirical study report about DSS on myocardial ischemia, so we selected DSS according to the original ratio of the dosage, carried out this research on it. Objectives: To observe the effects of DSS on experimental myocardial ischemia of animal, preliminarily explore the effective mechanism of DSS and offer experimental guideline with which CHD is prevented and treated clinically. Methods: 1 The effects of DSS on the rats'experimental myocardial ischemia induced by Isoprenaline injection (ISO): Fifty healthy male Wistar rats were divided into five groups at random, they were blank control group, model group, DSS low-dose group (raw herb 14g/kg·d) , DSS high-dose group (raw herb 28g/kg·d) and positive control group (FufangDanshenDiwan 0.081g/kg·d). Each group consisted of ten rats. The blank control group and the model group were fed with distilled water and the rest groups were fed with according medicine from the first day to the seventh day, once a day. From the fifth day to the seventh day during the experiment, injections of ISO (5mg/kg) were given to every group intraperitoneally except the blank control group to which injections of saline were given intraperitoneally one hour after taking medicine, so that the myocardial ischemia rats'models were duplicated. In the duration, we recorded the electrocardiogram of the tenth minute before the first injection of ISO and the thirtieth minute after the last injection of ISO. Two hours after the last injection of ISO, all the rats were killed, the blood were obtained and the activities of lactate dehydrogenase (LDH), glutamic oxalacetic transaminase (GOT) and creatine phosphokinase (CPK) were checked up. The apex of myocardium was cut and made into specimen of transmission electron microscope (TEM), observed the myocardium ultrastructure. The rest myocardium was made into 10% tissue homogenate with saline for checking up the activity ofsuperoxide dismutase (SOD) and the content of maleic dialdehyde (MDA). 2 The effects of DSS on the mice's experimental myocardial ischemia induced by pituitary injection (Pit): one hundred healthy male Kunming mice were divided into five groups at random, they were blank control group, model group, DSS low-dose group (raw herb 30g/kg·d) , DSS high-dose group (raw herb 60g/kg ·d) and positive control group (FufangDanshenDiwan 0.174g/kg·d), each group consisted of twenty mice. The blank control group and the model group were fed with distilled water and the rest groups were fed with according medicine from the first day to the fifth day, once a day. On the fifth day injections of Pit (30U/kg) were given to every group intraperitoneally except the blank control group to which injections of saline were given intraperitoneally one hour after taking medicine, so that the myocardial ischemia mice's models were duplicated. In the duration, ten mice were chosen from each group and their electrocardiograms of the tenth minute before the Pit injections and the fifth minute, fifteenth minute, twenty-fifth minute after the Pit injections were recorded. Thirty minutes after the Pit injections the other mice in every group were cut head, the blood were obtained and the contents of nitric oxide (NO) were checked up. The tip of myocardium was made into pathological specimen to observe the activities of myocardium endothelio nitric-oxide synthase (eNOS) by immunohistochemitry methods. The rest myocardium was madeinto 10% tissue homogenate with saline for checking up the activity of SOD and the content of MDA. Result: 1 The effects of DSS on the rats'experimental myocardial ischemia induced by ISO 1.1 The effects on the rats'electrocardiogram: The electrocardiographic abnormalities were taken as the positive reaction,such as the displacing of J-point (stepping up or down more than 0.1mv) or the abnormality of T-wave or the emergence of Q-wave, the positive animals were counted and the dates were analyzed. The results showed that no positive reaction took place in the blank control group while positive reaction took place in all the ten rats of the model group. There was significant difference between the two groups (P<0.01), showed that the models were successful. There were three rats showed positive reaction in the DSS high-dose group, four showed positive reaction in both the DSS low-dose group and the positive control group, compared with the model group, there were significant difference (P<0.05 or P<0.01). 1.2 The effects on LDH, GOT and CPK in serum: Compared with the blank control group, the contents of LDH, GOT and CPK in serum in the model group increased obviously (P<0.01), showed that the models were successful. Compared with the model group, the contents of LDH, GOT and CPK in serum in the DSS high-dose group, the low-dose group and the positive control group decreased obviously (P<0.05 or P<0.01).1.3 The effects on the rats'myocardium ultrastructure: Under the TEM,the result showed that in the blank control group, the cardiac muscle fibers were in good order and the mitochondria were integrated, the mitochondria cristae were regular. The model group's cardiac muscle fibers were disorder, while the mitochondria were damaged and the cristae were fused or disappeared. The DSS high-dose group's cardiac muscle fibers were regular basically, the mitochondria were intact and the cristae were fused or disappeared occasionally. The DSS low-dose group's cardiac muscle fibers were disorder slightly, the mitochondria were intact and cristae were fused or disappeared partly. The positive control group's pathologies were similar with the DSS low-dose group. 1.4 The effects on the activities of SOD and the contents of MDA in myocardium: Compared with the blank control group, the activities of SOD in the model group decreased obviously (P<0.01), while the contents of MDA in the model group increased obviously (P<0.01). Compared with the model group, the activities of SOD in the DSS high-dose group, the low-dose group and the positive control group increased significantly (P<0.05 or P<0.01), while the contents of MDA decreased significantly (P<0.05 or P<0.01). 2 The effects of DSS on the mice's experimental myocardial ischemia induced by Pit 2.1 The effects on the mice's heart rate: There wasn't significant difference in the mice's heart rate before injected Pit among allthe groups (P>0.05). In the fifth minute, the fifteenth minute and the twenty-fifth minute after injections of Pit, the heart rates in the model group decreased significantly compared with the blank control group (P<0.01), showed that the models were successful. The heart rates of the DSS high-dose, the low-dose group and the positive control group recovered significantly compared with the model group (P<0.01 or P<0.05). 2.2 The effects on the content of NO in the serum: Compared with the blank control group, the content of serum NO in the model group decreased obviously (P<0.01). Compared with the model group, the content of serum NO in the DSS high-dose group, the low-dose group and the positive control group increased significantly (P<0.01). 2.3 The effects on the activity of the myocardium's eNOS: Under the optical microscope,it showed that the activities of eNOS in the model group decreased compared with the blank control group, while the activities of eNOS in the DSS high-dose group, the low-dose group and the positive control group enhanced compared with the model group. 2.4 The effects on the activities of SOD and the contents of MDA in the mice's myocardium: Compared with the blank control group, the activities of SOD in the model group decreased obviously (P<0.01), while the contents of MDA in the model group increased obviously (P<0.01).
