Objective:To discuss the effect of XueFuZhuYu decotion for promoting angiogenesis and expound its mechanism by observing XueFuZhuYu decotion's effect for protecting ischemic myocardium of myocardial ischemia rat model.Methods:The left anterior descending coronary artery was ligated to make the model of myocardial ischemia in SD rats.They were divided into 4 gorups at random.These groups were the model group,Shexiangbaoxin pills group, XueFuZhuYu decoction group and the sham operation group.There were eight rats in each group.They were treated for 7 days by different therapies.After 7 days,all the rats were killed.Their blood and hearts were taken for some detections.We detected the content of SOD,MDA,NO and ET.We measured the myocardial infarction size.We studied the expression of CD34,VEGF at protein level with the immune histochemical method.We measured endothelial cells and the microvessel density in the edge of infarcted section to decide whether XueFuZhuYu decoction could induce myocardial angiogenesis and evaluate its molecular mechanisms.Results:The serum SOD,NO content of the model group was significantly lower than the sham operation group(P<0.01),while XueFuZhuYu decoction group, SheXiangBaoXin pills group were significantly higher than the model group(P<0.05).The XueFuZhuYu decoction group,SheXiangBaoXin pills group were significantly lower than the sham operation group(P<0.05).The XueFuZhuYu decoction group was higher than the SheXiangBaoXin pills group,but there was not significant difference(P>0.05).The serum MDA,ET content of the model group was significantly higher than the sham operation group(P<0.01),while XueFuZhuYu decoction group,SheXiangBaoXin pills group were significantly lower than the model group(P<0.05).The XueFuZhuYu decoction group,SheXiangBaoXin pills group were significantly higher than the sham operation group(P<0.05).The XueFuZhuYu decoction group was lower than the SheXiangBaoXin pills group,but there was not significant difference(P>0.05). The myocardial infarction size of the model group was bigger than the XueFuZhuYu decoction group,SheXiangBaoXin pills group(P<0.05),but the XueFuZhuYu decoction group and SheXiangBaoXin pills group were not significant difference(P>0.05). The endothelial cells and microvessel density of the model group were higher than the sham operation group(P<0.05).The endothelial cells and microvessel density of the treatment groups(XueFuZhuYu deccction group,SheXiangBaoXin pills group) were significantly higher than the model group(P<0.05).The treatment groups were significantly higher than the sham operation group(P<0.01).But compared with the XueFuZhuYu deccction group and SheXiangBaoXin pills group,there was no significant difference(P>0.05).The VEGF expression of the model group was significantly higher than the sham operation group(P<0.05),while XueFuZhuYu decoction group,SheXiangBaoXin pills group were significantly higher than the model group(P<0.05).The XueFuZhuYu decoction group,SheXiangBaoXin pills group were significantly higher than the sham operation group(P<0.01).The XueFuZhuYu decoction group was higher than the SheXiangBaoXin pills group,but there was not significant difference(P>0.05). Conclusion:The XueFuZhuYu decoction lowered serum MDA,ET and increased serum SOD, NO.The result showed that XueFuZhuYu decoction could resist oxidation and produce vasodilatation.At the same time it reduced the myocardial infarction size and increased microvessel density.The effect of its promoting angiogenesis was by upregulating VEGF expression.
