Experimental Study Of Wanyoujin Herb Serum's Effect On Th1/Th2 Cytokines In T Cells From Peripheral Blood Of Relapsing Condyloma Acuminatum Patients

Objective: It has been known that condyloma acuminatum(CA) is caused by human papilloma virus(HPV) infection which is a sexual transmited disease(STD). The high relapsing rate of CA patients is the main problem in clinical. It has been suggested that there is an imblance between Thl/Th2 cytokines during immune response to HPV and there is a clonal diversion towarads Th2 cytokines in the procedure that causes our immune system can not response effectfully and HPV can not be cleared. In the present study, we observed the effect of a traditional decoction, Wanyoujin, which is an experienced treatment for CA patients in our medical career, on Thl/Th2 cytokines secreated by T lymphyocytes derivied from CA patients peripheral blood samples in order to understand its mechanism to treat CA, that might provide theroy support for further clinical use of Wanyoujin decoction.Methods: Wanyoujin herb serum in mice were made, the serum were added to T cells during culture procedure, culture environment kept in 37, 5%CO 2 ?there were 4 groups in the experiment: Wanyoujin herb serum group, serum control group , CA control group and normal control group. Levels of cytokines interleukin-2(IL-2), interleukin-4 (IL-4)and interferon-y(IFN-y)were measured by ELISA method. The results were analysied in order to understand the herb serum's effect on the cytokines secreated by T cells.Results: in the first experiment, levels of IL-2, IL-4 and IFN-ybetween condyloma acuinatum patients and normal control group were assayed, the results showed a remarkedly decrease of IL-2 and IFN-ylevels and remarkedly increase of IL-4 level(p<0.05 or p<0.01 respectively). In the second experiment, the effect of Chinese medicia herbs decoction "Wanyoujin"on cytokines were studied.the results revealed that different doses(large, middle and small doses)could increase IL-2 and IFN-ylevels, decrease IL-4 level, however, the differences among three doses were not so significant, middle dose group had shown the best effect among three doses. In the last experiment, cytokines of IL-2, IL-4 and lFN-yamongWanyoujin group, serum control group and CA group were measured with the same methods.Results showed no significant difference between serum control group and CA group, however, when compared with Wanyoujin middle dose group,the differences were significant(p<0.05 orpO.Ol respectively).Conclusion: The present study reconformed a well-known theory that there was an imblance between Thl/Th2 cytokines during condyloma acuminatum infection which usually shows increase of Th2 type cytokine like IL-4 and decrease of Thl type cytokines like IL-2 and IFN-yet al.which means a weak Thl cells and relatively strong Th2 cells response during infection which is so called "clonal diversion". Our experiment revealed that Wanyoujin herb serum could regulate Thl/Th2 subgroups's imblance, increase Thl cytokines such as IFN-yand IL-2, strength body's immune function induced by Thl cells. Our results might explain from cytokine-cellular immune regulatory theory the mechanism of Wanyoujin treating relapsing condyloma acuminatum patients.
