| Objective:To investigate the effects of somatostatin and growth hormone on apoptosis of pancreatic acinar cells and apoptosis-regulated gene bax and bcl-2 in treating severe acute pancreatitis in rat.Methods:In sodium tauroglycocholate induced severe acute pancreatitis with or without treatment of somatostatin and growth hormone in rat, apoptosis of pancreatic acinar cells was detected by TUNEL method, and the expression of apoptosis-regulated gene bax and bcl-2 was determined by SABC immunohistochemical technique.Results:On TUNEL, the apoptotic index of pancreatic acinar cells in saline treated group was significantly lower than that of somatostatin treated group and somatostatin and growth hormone treated group ( P<0.05 ) . On immunohistochemistry, the postive rate of bax in somatostatin and growth hormone treated group was markedly higher than that of saline treated group (P<0.05) , but there was no marked difference on the postive rate of bax between somatostatin treated group and saline treated group; The postive rate of bcl-2 in saline treated group was markedly higher than that of somatostatintreated group and somatostatin and growth hormone treated group (P<0.05). Conclusion:Induction of apoptosis in pancreatic acinar cells injury might be one of mechanisms of somatostatin in treating severe acute pancreatitis in rat, and the mechanisms of apoptosis might be correlated with the down-regulation of Bcl-2, but has no relations with the up-regulation of Bax; Induction of apoptosis in pancreatic acinar cells injury might be one of mechanisms of somatostatin and growth hormone in treating severe acute pancreatitis in rat, and the mechanisms of apoptosis not only might be correlated with the down-regulation of Bax, but also has relations with the up-regulation of Bcl-2. |
