Objective:This experiment on rats with severe acute pancreatitis model,application of somatostatin combined with growth hormone to its intervention,comparison group,somatostatin and combined groups in all three groups the difference evaluation index,which somatostatin Joint growth hormone on severe acute pancreatitis (SAP) pancreatic key enzymes in apoptosis and Caspase-3 expression。Methods:1,Modeling:Bile duct injection of sodium taurocholate retrograde rat model of severe acute pancreatitis;2,Serum amylase:Automatic biochemical analyzer serum amylase;3,Detection of Caspase-3 protein expression:Detected by SABC pancreatic tissue expression of Caspase-3;4,Detection of apoptotic cells:TUNEL method, apoptosis of pancreatic acinar cells in each group were detected, counted 500 acinar cells the percentage of positive cells,ie apoptotic cells;5,Pathological scores of pancreas:HE staining of pancreatic organizations,according to Kusser standard score。Results:1,Successfully produced a rat model of severe acute pancreatitis;2,Serum amylase somatostatin than in the control group (P <0.05),serum amylase combination group was lower than that of somatostatin (P <0.05);3,Somatostatin group,the expression of Caspase-3 than the control group (P<0.05),the joint group of the expression of Caspase-3 group compared with the high somatostatin (P <0.05);4,Somatostatin group,apoptotic index of pancreatic cells than the control group (P <0.05), the Joint Group of pancreatic cell apoptosis index higher than somatostatin group (P <0.05);5,Somatostatin group,apoptotic index of pancreatic cells than the control group (P <0.05), the Joint Group of pancreatic cell apoptosis index higher than somatostatin group (P <0.05);6,Apoptotic index was negatively correlated with the pathological score (r =- 0.812, P <0.001)。Conclusion: 1,Somatostatin combined with growth hormone can promote apoptosis of pancreatic cells,increased pancreatic expression of Caspase-3 protein to reduce the pancreatic pathological damage,and better than a single application of somatostatin;2,Joint growth hormone somatostatin by upregulating the expression of Caspase-3 to promote apoptosis of pancreatic cells,reduce the pathological injury of the pancreas;3,Somatostatin combined with growth hormone treatment of rats with severe acute pancreatitis can significantly improve the pancreatic tissue morphology and reduce serum amylase levels,and better than the single application of somatostatin。...
