Objective: To explore the expressive state of TNF-α, Caspase-3 antigen and NF-κBmRNA in liver tissues of hepatitis patients and its significance in hepatocyte apoptosis. Methods: Sixty-five paraffin setions from patients with viral infection were studied and 10 cases of normal were as control. Immunohistochemistry was employed to determined the expression of TNF-α , Caspase-3 antigen; NF-κBmRNA was measured by in situ hybridization; Apoptosis was detected by terminal deoxynu— cleotidyl transferase mediated DUTP nick end labeling (TUNEL). Results: (1)The positive expression rate of TNF-α, Caspase-3 antigen and NF-κBmRNA in patients with viral hepatitis was 76. 9%, 69.2%, 70. 3% respectively; The positive rate in control was 20%> 20%, 10% respectively, they showed great significant difference (P<0. 005). (2) The positive stain of TNF-α , Caspase-3 antigen and NF- k BmRNA in viral hepatitis with hepatocyte apoptosis was 86. 0%, 79.1%, 79. 1% respectively; and the positive stain in no hepatocyte apoptosis was 59. 1%, 50%, 54.5% respectively, and the difference were significant (P< 0.05). (2) The more severe the disease, the higher theexpression of TNF-α ,Caspase-3 antigen and NF- κBmRNA, but the expression of TNF-α had no significence (P>0. 05). (3) There were highly correlations among TNF- α,Caspase-3 antigen and NF-κ BmRNA in liver tissue of hepatitis (P<0. 05). Conclusion: (1)TNF- α ,Caspase-3 antigen and NF- κ BmRNA played an important role in viral hepatitis occurrence. (2) TNF- α, Caspase-3 antigen and NF- κ BmRNA were related with viral hepatitis occurrence, advance and hepatocyte apoptosis. (3) Though the expression of TNF- a antigen had no significent, it reflected the characters of expression. Cases should be enlarged to observed in the future. 4)Combined detection of the above four can reflect degree of hepatocyte damage, which can give new measures to prevent from the development of hepatocyte apoptosis and liver f ibrosis.
