| objective transforming growth factor signaling pathway is involved in a variety of important cellular regulative functions.including cell growth, differentiation ,adhesion migration .extracellular matrix formation and immune regulation. The study was aimed to evaluate the relationship and mechanism of transforming growth factor 1 protein and smad4mRNA in formation and development of fibrosis of viral hepatitis by detect expression of transforming growth factor 1 protein and smad4mRNA in liver biopsy and assay serum HA, LN, C-IV. Methods in situ hybridization and immunohistochemistry was used in 10 cases of normal liver tissue, 14 cases of liver cancer , 60 cases of chronic viral hepatitis and cirrhosis; enayme-linked immunoquantitative method was uaed in 62 cases of serum HA, LN, C-IV .Results Increased expression of TGF- 1 and smad4mRNA in the chronic viral hepatitis and cirrhosis; the positive rates ofsmad4mRNA were obviously higher in chronic viral hepatitis and cirrhosis (72.5%,80%) than in the normal liver (30%) and the liver cancer (28.6%), the expression of smad4mRNA was a significant relationship between the viral hepatitis, the normal liver and the liver cancer(P<0.05); most of the positive cell were distributed surroundings of portal tract,central vein,sinus.Conclusions The expression of TGF- 1 and smad4 showed positive correlation each other and showed intimate relationship in different stage of fibrosis of viral hepatitis; TGF- /smad pathway have a significance in formation and development of fibrosis of viral hepatitis. |
