Objectives:This research was designed to investigate the expression and relationship of PARP-1 and caspase-3 protein in normal breast, benign lesional and breast cancer, and their correlation with clincopathologic features, for provide evidence for breast cancer of diagnosis and prognostic evaluation.Methods:1 The 282 paraffin-embedded specimens were collected, including 267 cases of invasive breast cancer and 15 cases of benign lesions. 11 cases of fresh normal breast, 10 cases of fresh benign lesions and 10 cases of fresh invasive breast cancer were collected. Diagnosis of all cases, sub-type classification, according to the WHO(2003) breast tumor histologic classification.2 The expression of PARP-1 and caspase-3 was measured by immunohistochemistry SP methods in 11 cases of normal breast, 25 cases of benign lesions and 277 cases of invasive breast cancer. Analyze the relationship between the expression of PARP-1 and caspase-3 and the pathological characteristics.3 In situ hybridization method was used to examine for the expression, relation and the effect of PARP-1 mRNA in 10 cases of fresh normal breast, 10 cases of fresh benign lesions and 10 cases of fresh invasive breast cancer.Results:Immunohistochemistry results showed that1 The expression and relevance of the PARP-1 and caspase-3 protein1.1 The expression of the PARP-1 protein in breast cancer, benign breast lesions and normal breast The positive rate of the expression of PARP-1 in breast cancer (65.70%, 182/277) is higher than that in normal breast (27.27%, 3/11) and benign lesions(44.00%, 10/25), the difference has statistical significance(p=0.009, p=0.011). Increased expression of PARP-1 may be related to the malignant biological behavior of breast cancer. PARP-1 and caspase-3 were conduced to the judgement and prognosis of breast cancer.
