Objective: To study the relation of Fas, Caspase-3 , NF- κ B and cardiac muscle cell apoptosis ,in order to know the machenism of myocardial function and the role of Fas ,Caspase-3 , NF-κ B in signal transduction in septic rats. Methods:30 Wistar rats were randomly divided into control group(n=6),septic group(n=24).The septic group was divided into 4 groups of 2h , 3h, 4h , 5h after cecal ligation and puncture(CLP).Myocardial tissue Fas,Caspase-3 ,NF-κ B protein levels and cardiac muscle cell apoptosis were measured at various intervals.At the same time myocardial function were also determined .Results: Fas,Caspase-3, NF-KB protein levels at 2h, 3h were significantly elevated compared to the control group in myocardial tissue (p<0.01 ).but there was not a noble discrepancy between 4h and 5h(p>0.05).The number of cardiac muscle cell apoptosis was higher in CLP than the control group(p<0.01) . The number of cardiac muscle cell apoptosis at 2h and 3h was higher than at 4h and 5h, there was a noble discrepancy (p<0.05).At first the myocardial function decreased, and then increased .LVSP and ±dp/dtmax were less at 2h and 3h compared to the control group(p<0.01),but there was not a noble discrepancy between the control group and 4h,5h(p>0.05),which was a noble discrepancy between 2h, 3h and 4h, 5h(p<0.01). Under the electron microscope ,different morphologicaldisorders of cardiac muscle cell were observed,mitochondria swelled. Conclusions:Levels of myocardial tissue Fas , Caspase, NF-κB were higher significantly at 2h, 3h,while were less at 4h,5h ,which led to cardiac muscle cell apoptosis through signal transduction in CLP-induced spepsis. The changes of heart function was relevant to cardiac muscle cell apoptosis. The above results showed that cardiac muscle cell apoptosis was relevant to levels of Fas , Caspase-3 , NF- κ B and led to myocardial injury.
