Protection Of Ozagrel Sodium Injection To Myocardial Cell Of The Myocardial Ischemia Model In Rat

Objective To discuss the possible myocardial protection of Sodium Ozagrel (OZG) injection to mycardial ischemia model in Wister rats.Method Forty healthy Wistar rats were selected and divided into five groups of eight rats each at random.They were blank control(group A),model(group B), pretreatment(group C),treatment(group D), affiliation with angised (group E). Every groups were fed with common fodder. The rats in Group A served as blank control whereas rats in Group B were given daily intraperitoneal injection of 2.0 ml isotonic Na chloride.Rats in Group C received OZG in the doses of 33.67mg/kg, Tow week later, apart from Group A, the other groups were given intraperitoneal or subcutaneous injection of more isoprenline hydrochloride(10mg/kg) produced myocardial ischemia model. Based on the depiction of ECG, ST-segment elevation≥0.1mv rats for modeling success. Group D and Group E in the modeling success after five minutes, were given diluted OZG(33.67mg/kg), OZG(33.67mg/kg) + nitroglycerin (0.17mg/kg) intraperitoneal injection. Depiction of every groups after modeling success within five minutes of ECG.. After one hour, with 10% Chloral Hydrate(0.3ml/100mg) intraperitoneal injection of anesthesia rats in each group, blood was from the abdominal aorta and centrifugal separation to be left out of the serum CK, LDH, AST, SOD, MDA. At the same time, from myocardial with quick-frozen in liquid nitrogen preservation, were used immunohistochemical methods in the detection of myocardial cells of caspase-3, caspase-9, Bcl-2 and Bax protein expression.Result ECG:Use large doses isoprenaline hydrochloride (10mg/kg) production model of myocardial ischemia, Each group of rats with myocardial ischemia leadde ECG II of ST-segment elevation were≥0.1mv. Every group's ST- segment raising range of II compared with itself group ST- segment raising range of II after 5 minutes , Group C,D and E all have significant difference( P<0.05). Compared with Group B's ST- segment raising range of II of the modeling success after 5 minutes, it is significant difference in Group C,D and E( P<0.05). Compared with Group E's ST- segment raising range of II of the modeling success after 5 minutes, it is significant difference in Group C and D(P<0.05). Orrhology detection:Compared with group A, the levels of AST, LDH, CK, MDA and SOD in serum Group B have significence difference (P<0.05).Compared with the levels of LDH, CK and AST of Group B, there are significant differenc in Group C, D and E (P<0.05). Compared with the level of MDA of B group, there are significant difference in Groups C and E(P<0.05). But there is not significant difference in Group C and E( P<0.05). Compared with the level of SOD of Group B, there are significant difference in Group C and E( P<0.05). Compared with the level of SOD of Group D, there are significant difference in Group C and E. Immunohistochemical result Compared with caspase-3,Bcl-2,Bax protein of Group A, the other four groups all have significant difference( P<0.05). Compared with caspase-3 protein of Group C, there are significant difference in Group D and E(P<0.05). Compared with caspase-9 protein of B group, there are significant difference in Group C,D and E( P<0.05). Compared with caspase-9 protein of Group D, there are significant difference in Group C and E( P<0.05). Compared with Bcl-2 protein of Group C, there are significant difference in Group B,D and E (P<0.05).Compared with Bax protein of Group E, there are significant difference in Group B,C and D(P<0.05). But there is no significant difference between Group C and E (P<0.05). Conclusion A great quantity of isoprenaline hydrochloride (10mg/㎏) could make well model of myocardial ischemia of rat by the way of hypodermic injection or intraperitoneal injection .Sodium ozagrel injection could reduce the level of free competition in the acute myocardial ischemia of rat ,and alleviate the pain of convulsionary coronary artery and angina pectoris .It has influencing in apoptosis of cardiac muscle cells in model of myocardial ischemia of rat .It could inhibited the protein expression of caspase kindred ,Bcl-2 and Bax in order to inhibite the apoptosis .accordingly Sodium ozagrel injection could protect the cardiac muscle cells in model of myocardial ischemia of rat...
