Objective:To investigate the concentration of serum HGF and the expression of HGF in kidney of type 2 diabetic rat model in different stages,and to study the effect in the pathogenesis of diabetic nephropathy (DN).Methods:Type 2 diabetic rat models were induced by high-fat and high-glucose diets and intraperitoneal injection with low dose streptozocin (30mg/kg).The expression of HGF in kidney were observed in different stages by immunochemistry. At the same time, blood glucose,insulin, cholesterol, triglyceride,HGF and 24-hour urinary protein were measured.Result:The concentration and the expression of HGF in kidney of diabetic group were significantly increased( P<0.01).Its level were markedly increased in the early stage and obviously decreased in the late stage.Conclusion:HGF may play an important role in the pathogenesis of diabetic nephropathy. The increase of serum HGF in the early stage seems to be an early index of DN.The dynamic change of HGF can predict the progress of DN. The decrease of HGF in kidney in the late stage appears to be a risk facter of DN severity.
