Objective: Through the comparative study of part HLA classⅡgene in Shanxi region between the gestational diabetes mellitus(GDM) and normal pregnancy group,we found the susceptibility genes and protective genes of GDM in Shanxi region. And to suggest the susceptible group prevent and intervent earlier, avoiding the occurrence of GDM and the type 2 diabetes.Methods: 39 cases of GDM and 42 cases of normal pregnant women were enrolled in this study,who were excluded diseases correlated with HLA gene in the first hospital of shanxi medical university from 2006.3 through 2007.3. 81 blood samples were collected in vacutainer tubes with EDTA from pregnant women, 39 with GDM and 42 without GDM .Then the samples were frozen .in -70℃refrigerator; Saturated phenol - chloroform extraction method of genomic DNA was used; Part sequence of HLA-II gene was examined by polymerase chain reaction sequence specific primer(PCR-SSP). Based on the actual laboratory work and requirements of statistical analysis , 30 cases in GDM group and 30 cases in control group sent to sequencing. Statistical analysis was performed using SPSS statistical software, the groups were compared usingχ2 test , the results were considered significant at P<0.05.Results:(1)Significant differences were noted between the two groups in HLA-II gene frequency in Shanxi region,which were consistent with previous reports; (2)It was observed that gene frequency of HLA-DRB1*0301,DQB1*0201 was increased in GDM group compared with the control group (38.5%vs16.7%;28.2%vs9.5%), there was significant difference between two group. (P﹤0.05). (3)We also observed a decrease in HLA-DQA1*0103 was seen in GDM(7.7%vs14.3%),there was also significant difference between them. (P﹤0.05). (4) The gene frequency of HLA- DQA1*0501 was increased in GDM group compared with control group, but the statistical difference was not significant(P﹥0.05).Conclusion:HLA-DRB1*0301 and DQB1*0201 gene are the susceptible genes of GDM in Shanxi region. HLA-DRA1*0103 may be the protective genes of GDM in this area.
