Experimental And Clinical Studies On Correlation Of Erzhitiangui Decoction's Improving The Quality Of Oocytes And Insulin-like Growth Factor-1 R MRNA Expression In Granulosa Cell

Objective: To observe the effect of Erzhitiangui(EZHTG) decoction on improving the quality of oocytes, and explore its functional mechanism. Methods: Experimental study: 60 mice were randomly divided into four groups: the first herb group(treated with EZHTG decoction and HMG) and the first control group(treated with HMG) were observed about the number of mature oocytes, After that, IEM was performed on the granulosa cell to observe the IGF-lR's location in granulosa cell. In situ hybridization was performed on the second herb group(treated with EZHTG decoction and HMG) and the second control group(treated with only HMG) to observe the expression of IGF-1R m RNA. Clinical study: 57 patients of IVF were randomly divided into two groups: the herb group(treated with EZHTG decoction combined with western medicine) and the control group(treated with western medicine). Symptoms of renal asthenia before and after treatment, the ratio of mature oocytes, fertilization, cleavage and pregnancy were observed. Results: Experimental study indicated that the ratio of mature oocytes of the first herb group is markedly higher than the control group (P<0.05) . IGF-1R locates on the membrane of lysosome in granulose cell. The quantity expression of IGF-lRm RNA in granulose cell of the second herb group is higher than the second control group (P<0.05) .The ratio of mature oocytes of mice positively correlates with the quantity expression of IGF-lRm RNA in granulose cell (P<0.01) .Clinical study indicated that the herb group was markedly better than the control group in ameliorating the symptom of renal asthenia and improving the ratio of mature oocytes , fertilization, cleavage and pregnancy (P<0.05)o Conclusion: IGF-1R locates on the membrane of lysosome in granulose cell. EZHTG decoction's function of improving quality of oocyte may relate to molecular function of IGF-1.EZHTG decoction can improve the quality of oocyte , and then increase the ratio of fertilization, cleavage and pregnancy.
